ObjectiveStudy on the alcohol extract of Polygonum perfoliatum L parts in vitro on herpes simplex virus(HSV-1)inhibition;optimize the extraction process of flavonoids in Polygonum perfoliatum L.preparation of Polygonum perfoliatum L anti herpes virus gel;study the smartweed quality standards of anti herpes virus gel.MethodThe CPE(Cytopathogenic effect,CPE)and thiazole blue(MTT)colorimetric method for the alcohol extract of Polygonum perfoliatum L were HSV-I in vitro antiviral experiment,to evaluate the alcohol extract of Polygonum perfoliatum in vitro anti HSV activity of parts;in order to extract flavonoids of Polygonum perfoliatum L.rate as the index,the best extraction process of single factor test and orthogonal test of Polygonum perfoliatum L.;with multi index evaluation,the orthogonal test was used to optimize the prescription Polygonum perfoliatum L anti herpes virus gel;the quality standard of Polygonum perfoliatum L anti herpes virus gel was determined by TLC,stability test and HPLC.ResultThe results of CPE indicated that it could completely inhibit HSV-I at low concentration of 0.78 mg·m L^-1.MTT displayed on this concentration of the virus inhibitory rate of 40%;the extraction process of the Polygonum perfoliatum L is optimized as follows:80%ethanol as solvent,the extraction temperature of,the solid to liquid ratio of 1:10,extraction of 2 times,each time for 2 h;the prescription of anti herpes virus gel is Polygonum perfoliatum L extract 0.13%,carbomer940 1%,glycerol5%,propylene glycol 5%,ethanol 3%,4-hydroxyethyl ester 0.2%;the obtained anti herpes virus gell was brown transparent gel,gas micro,easy to apply;p H,particle size and so on in line with Pharmacopoeia;no skin irritation;good stability.In this study,a stable and reliable method was established for the determination of the Polygonum perfoliatum L anti herpes virus gel with the rutin as the standard.ConclusionIn this experiment,the anti herpes virus test was carried out by adding alcohol extract of Polygonum perfoliatum L;the result showed that the Polygonum perfoliatum L was a low toxicity and effective anti herpes virus drug;the alcohol extract of Polygonum perfoliatum L was made into the gelling agent;the quality standard of Polygonum perfoliatum L anti herpes virus gel,this study provides a basis for further research on anti herpes virus gel.
