Objective Determined the effective part of Polygonatum odoratum on nourishing Yin and inspected the fundamental properties of different processed Polygonatum odoratum.Used fingerprint and Yin-deficiency rat to compare the quality of different processed Polygonatum odoratum and established the best integrated processing method of crude Polygonatum odoratum in original planting area.Methods Ethanol was used to divide the water extraction of Polygonatum odoratum into three fractions(? aqueous part ? ethanol part ?precipitation part).T3 was used to establish Yin-deficiency rat model.ELISA method to detect the content of FT3,FT4 in serum and cAMP,cGMP in plasma.Tested the content of water,total ash,extract and polysaccharides of different processed Polygonatum odoratum in accordance with the requirement of pharmacopoeia.Using HPLC to establish fingerprint of the active fraction of different processed Polygonatum odoratum,evaluated the fingerprint similarity and methodology.Results All fractions can increase the weight,reduce the temperature and regulate the level of FT3,FT4,cAMP,cGMP in the model rat,its effect had significant difference(P<0.01)and the part ? was the best.All processed Polygonatum odoratum fulfilled the requirement of pharmacopoeia.By studying comparatively the fingerprint of 21 samples,13 common peaks had been confirmed and the similarity were high(>0.8).The method for making fingerprint was stable and reliable.There were no significant differences between the simple treatment samples and the processed samples.Processing process can not enhance the effect of nourishing Yin of Polygonatum odoratum significantly.Conclusion Polygonatum odoratum had certain therapeutic effect on Yin-deficiency rat induced by T3 and the part ? was the best.The pharmacopoeia standard and fingerprint can not distinguish the difference between various processed samples accurately and there were not much difference in compositions and efficacy between them.So the best integrated processing method of crude Polygonatum odoratum in original planting area that we recommended was to slice and dry simply.
