The property and flavor of Polygonatum Odoratum is sweet taste and neutral nature.It is frequently used to nourishing yin,moisturizing lung and promoting the production of body fluid.Although its pharmacodynamic action is moderate,but will not hold the pathogenic factor.Polygonatum Odoratum is a principal drug of some famous prescription,such as Jiajianweirui-decoction, Yuzhumaidong- decoction and Yiwei- decoction,so it has good drug action. Because of the pharmacodynamic action,alimentary action and easily growth, deeply study on Polygonatum Odoratum will bring most major social effects and economic returns,it is worth to pay close attention to the Polygonatum Odoratum.Objective:This thesis adopt strategy of investigating active components new drug,and use means of literature study and empirical study to research the antiaging action of Polygonatum Odoratum,in order to achieve the followobjectives:1.Used the chemical methods and pharmacodynamics screening technique in order to elucidate the antiaging active components of Polygonatum Odoratum.2.To explore a pharmacodynamics bolting system that is convenient, easily,cheapand effective.In order to research the mechanism of Polygonatum Odoratum anti-aging by multisystem,multistrata,multi-angular angle.3.To study and research the quality criteria of antiaging active components of Polygonatum Odoratum preliminarily in order to establish a quality criteria system for Polygonatum Odoratum.4.Research the mechanism of Polygonatum Odoratum anti-aging by chemical methods and pharmacodynamics screening technique.And then,provide experical basic for exploitation of anti-aging new drugs in Polygonatum odoratum.Methods:This thesis contained tow parts of literature study and empirical study. The part of literature study mainly analysis the present situation and perspective of Polygonatum odoratum product research and development,at the same time overview the modern mechanism of antiaging of Chinese herb medicine, the principal antidotal active component and the present situation of aging model and study methods of aging new drugs.So that to establish a strategy of investigating for empirical study.The part of empirical study observe the anti-aging effct of aqueous extract of Polygonatum odoratum firstly,and explore the feasible and representation of pharmacodynamics bolting system.Then according to magnitude of polarity of solvent to separate the Polygonatum odoratum in five parts.Observe each parts antiaging action by the bolting system that explored by the above experiment.Choose the best part as the antiaging active parts of Polygonatum odoratum.Qualitation and quantitative of antiaging active parts is analyzed.The spectrum of a graph of HPLC is detected also.On account of that result,antiaging active parts is separated in 3 components.Each components' antiaging action also be observed by the pharmacodynamics bolting system.To provide experical basic for explanation of mechanism and quality criteria in antiaging active component of Polygonatum odoratum,All in a11,the whole part of empirical study apply pharmacodynamics bolting system all the time and use the chemic method to purify Polygonatum odoratum,in order to elucidate the antiaging active components and provide experical basic for exploitation of anti-aging new drugs in Polygonatum odoratum.Result:1.Polygonatum odoratum extracts has good anti-aging effects,with low-dose an antioxidant,play a significant role on removing free radicals, and high-dose evidently regulating function in immune system and nervous system.2.From vitro and vivo experiments,anti-oxidation and removal of radical system,immune endocrine system and nervous system,DPPH clearance rate,the total reduction capacity,memory of learning,endurance of swimming,index of immune organs and sexual organs,pathological biopsy of Thymus and spleen, liver tissue MDA and plasma T-SOD,all these 11 indicators formatted the anti-aging screening system of Pharmacodynamics that can fully reflect the general pathology of aging and the application has certain feasibility and representative.3.Compared with other polar fraction,Water extracted fraction that use system solvent method has better anti-aging effects,and more conspicuous impact on every index in this system.So it is presumed to be the effective parts of anti-aging.4.Systematic chemical assay and quantitative analysis of the effective fraction of Polygonatum odoratum showed that it main contains polyose(54.17%), flavonoids(1.27%),saponins(12.48%)as well as protein and tannic acid. Studying the assaying methods on all composition,it manifested that the assaying system,ultraviolet - visible spectrophotometry combined with other Color Methods,is simple,economical and reliable,and provides foundation for quality control.The determination of the total count of every composition can be more accurate and avoid mutual interference by using this method.5.When the chromatography conditions are as follows:column:Dikma Diamonsil C185μm 250×4.6mm;flow rate:1.00 ml/min;sample volume:25μl; detect W/L:300 nm;column temperature:room temperature;mobile phase:VH20: VMeOH:VAN= 2:3:3,HPLC chromatogram,reflect more peak information,can be basi-phenogram of effective anti-aging fraction of Polygonatum odoratum and guidance of quality control.6.The effective anti-aging fraction has been further separated by column chromatography into three parts,named polysaccharide fraction(89.64%), flavonoids fraction(76.92%)and the more complex remaining parts.Comparing with the pharmacodynamics effect of the three parts in isodose condition,it shows that flavonoids fraction has better function in vitro antioxidant,the improvement of plasma T-SOD activity and effect on content of MDA in liver tissue are obviously surpass the other two,while polysaccharide fraction surpass the others on endurance,memory,thymus and spleen index of the aging mice.Conclusion:We had initially found the Polygonatum odoratum anti-aging active components,And conducted the function mechanism and the quality specification,by using modern medicine researching methods and train of thought.The result indicated that Polygonatum odoratum as the traditional herbs have the good antiaging function,by chemistry separation and pharmacodynamics tracking system.And the active components which withdraws through the system solvent extraction,more active under the same level dose surpasses,Simultaneously the polysaccharide class,anthoxanthin class and saponin class and so on slightly.The big active components are obvious: the polysaccharide class and the anthoxanthin class ingredient function,But their functions mechanism are different respectively,obviously the active components are under these two ingredient synergism result.Therefore,the anti-aging active components in Polygonatum odoratum should be take the water-soluble polysaccharide and the anthoxanthin glucoside as the main active constituents,and both components which mix mutually according to certain proportion,Its function mechanism is possible that the oxidation resistance elimination free radical,the adjustment immunity internal secretion and the adjustment nervous system and so on.Uses the ultraviolet - visible light spectrophotometric method and unifies the different colored method the content determination method system as well as the highly effective liquid phase atlas attribute description and so on can initially carry on the monitoring to the effective spot quality.The anti-oxidation activity of aqueous extract of Polygonatum odoratum as a new medicine is worth further developing,this study has provided certain experimental foundation for it.
