Objective: To explore the effect of Shennao Fuyuan Decoction combined with hUC-MSCs transplantation on the expression of Nogo-A and GAP-43 in MCAO rats.Methods: Fifty SD rats were randomly divided into sham operation group and model preparation group by random number table method.The model preparation group used the suture method to prepare the middle cerebral artery ischemia-reperfusion model,and then randomly divided into the model group,the Shennao Fuyuan Decoction group,the hUC-MSCs group and the Shennao Fuyuan Decoction combined with hUC-MSCs group.Before treatment and after treatment,each rat was scored with m-NSS.The rats were sacrificed on the 3rd day after the operation.The pathological changes of brain tissue were observed by HE staining,Nogo-A and GAP-43 protein expression in brain tissue were detected by immunohistochemistry,and Nogo-A and GAP-43 mRNA expression in brain tissue were detected by q RT-PCR.Results: 1.Behavioral changes in rats: Shennao Fuyuan Decoction combined with hUC-MSCs group can significantly elevate the symptoms of neurological deficits in MACO rats.2.HE staining:The structure of nerve cells in the sham operation group is normal,with regular arrangement and clear outlines.After cerebral ischemia,nerve cells in the model group decreased,a large number of nerve cells were degenerated,cell structure was loose,arranged disorderly,nuclei were unclear,shrunk or appeared cavities,and some nerve cells died.Compared with the model group,the pathological changes of the Shennao Fuyuan Decoction group,hUC-MSCs group and the combination group were reduced,the number of nerve cells increased,the amount of necrosis was reduced,the cell structure was denser,and the outline was clearer.The pathological changes in hUC-MSCs group were lighter.3.Immunohistochemical detection: Low signal Nogo-A and GAP-43 expressions were seen in the sham operation group.Nogo-A and GAP-43 protein in each group were higher than those in the sham operation group(P<0.05).Compared with the model group,Nogo-A protein was decreased in Shennao Fuyuan Decoction group,hUC-MSCs group and combination group,and GAP-43 protein was increased(P<0.05).Compared with the combination group,Nogo-A protein was higher in Shennao Fuyuan Decoction group and hUC-MSCs group,and GAP-43 protein was lower(P<0.05).4.q RT-PCR detection: Compared with the sham operation group,Nogo-A and GAP-43 mRNA in the model preparation group increased(P<0.05).Compared with the model group,Nogo-A mRNA was weakened in Shennao Fuyuan Decoction group,hUC-MSCs group and combination group,and GAP-43 mRNA was enhanced(P<0.05).Compared with the combination group,Nogo-A mRNA was enhanced in Shennao Fuyuan Decoction group and hUC-MSCs group,and GAP-43 mRNA was weakened(P<0.05).Conclusion: 1.After ischemic stroke in rats,Shennao Fuyuan Decoction combined with hUC-MSCs transplantation can alleviate the performance of neurological deficits in rats and reduce the scores of neurological deficits in rats.2.Shennao Fuyuan Decoction combined with hUC-MSCs transplantation may be able to regulate the expression of GAP-43 and Nogo-A protein,up-regulate the expression of GAP-43 mRNA,down-regulate the expression of Nogo-A mRNA,and play a neuroprotective effect.
