Objective:To investigate the effects of Shennao Fuyuan Decoction on cerebral angiogenesis and integrin ? 5 ? 1 expression in rats with ischemia-reperfusion.Methods:120 SPF male Sprague-Dawley rats were selected.According to the principle of randomized control,30 were selected as blank group and 30 were sham-operated group.The other 60 rats were used to establish the middle cerebral artery occlusion(MCAO)model.After successful modeling,they were randomly divided into model group and traditional Chinese medicine group.Each component is three phases of 3d,7d,and 14 d.The blank group,the sham operation group,and the model group were given the same volume of normal distilled water,and the Chinese medicine group was given the same volume of kidney-recovery soup.The m-NSS score was used to evaluate the degree of neurological deficit in rats.Hematoxylin and eosin(HE)staining was used to observe the morphological changes of brain tissue cells at different time points.Immunohistochemistry was used to determine MVD.Western Blot was used to detect the expression of integrin ? 5? 1 in cerebral ischemic tissue.Happening.The experimental data were expressed as mean ± standard deviation(`x ± s).The LSD method was applied when the two groups were compared and the variance was homogeneous.The Games-Howell method was applied when the variance was not uniform.p<0.05 suggested that the difference was statistically significant.Results:1.Neurological function score: There was no neurological impairment in the blank group or the sham operation group,and the m-NSS score was 0.In the model group and the Chinese medicine group,there were significant neurological impairment symptoms(p<0.05)in the blank and sham-operated groups,suggesting that successful modeling resulted in ischemic injury to the brain tissue and neurological disorders.In the same phase,the scores of the Chinese medicine group were lower than those of the model group,and the difference was the highest at 14 days(p<0.05),suggesting that the Chinese medicine group can promote the recovery of nerves after cerebralinfarction in rats to a certain extent,and the duration of treatment Longer.2.HE staining: no obvious damage was observed in the blank group and sham-operated cortical neurons.The cell structure was intact,the arrangement was regular,and the staining was uniform.At the same time,the structure of the nerve cells around the ischemic area of the model group was destroyed,cell edema,vacuolization,cell shrinkage,nucleus pyknosis,chromatin edge collection,darkening,uneven staining,and disordered arrangement.On the 3rd day after modeling,the morphological changes of neuronal cells in the Chinese medicine group were similar to those in the model group.On the 7th and 14 th day after modeling,the neuronal cell degeneration of the traditional Chinese medicine group was reduced compared with the model group,and the arrangement was relatively neat.The cell edema,vacuolization and nuclear pyknosis were significantly reduced.3.Microvessel density: A small amount of neovascularization was seen in the blank group and the sham operation group.At different times,the MVD of the model group and the Chinese medicine group was significantly increased compared with the first two groups.It can be seen from the analysis data that the MVD peaked at 7 days after MCAO modeling,and increased at 3d-7d and decreased at 7d-14 d.Compared with the model group,the MVD of the traditional Chinese medicine group increased significantly(p<0.05),suggesting that Chinese medicine has a certain up-regulation effect on angiogenesis.4.Integrin? 5? 1 expression: The positive expression rate of each group was lower in the blank group and the sham operation group.The positive expression of integrin ? 5 ? 1 was significantly increased in the model group and the Chinese medicine group(p<0.05),indicating success.Expression of ? 5 ? 1 can be induced after modeling.The positive expression of ? 5 ? 1 in each group gradually increased with time at different phases.At the same time,the expression of ? 5? 1 in the Chinese medicine group was more than that in the model group(p<0.05),indicating that the Chinese medicine can regulate the expression of? 5? 1.Conclusions:After cerebral ischemia in rats,Shennao Fuyuan Decoction can significantly improve the neurological deficit in rats,which increase theexpression of integrin ? 5? 1 in rat brain tissue and promotethe increase of vascular density.
