Interventional Effect Of Jiawei Baoyuan Decoction On ACE2-Ang(1-7)-MAS Axis And MiR-1 In Rats With Heart Failure

Objective: To observe the regulation of Jiawei Baoyuan Decoction on ACE2-Ang(1-7)-MAS axis and mi R-1 in rats with heart failure.Methods: 60 rats were divided into blank group,control group,traditional Chinese medicine group,and positive control group,with 15 rats in each group.Except for the blank group,the other three groups were modeled with isoproterenol.After successful modeling,the blank group and the model group were given saline by gavage,the test group was gavage with Jiawei Baoyuan Decoction,and the positive control group was gavage with valsartan.Three months later,the cardiac color Doppler ultrasound was observed;the contents of BNP,ACE2,Ang(1-7)and Mas in rat plasma were observed by immunohistochemistry;the mi R-1 of rat myocardium was detected by Real-time PCR and the pathological section was observed by MASSON staining.result:(1)Echocardiography: Compared with the blank group,LVEF and LVFS of the model group were decreased,while LVSD and LVDD were increased.The difference was statistically significant(p<0.05).Combined with the general situation,it indicated that the model was successful.Compared with the model group,the LVEF and LVFS of the traditional Chinese medicine group and the positive drug group increased,while the LVSD and LVDD decreased.The difference was statistically significant(p<0.05),indicating that both Jiawei Baoyuan Decoction and valsartan treatment can reduce left ventricular diastole It can improve the ejection fraction during the period and systole period.And there is no significant difference between the two effects(p>0.05).(2)Serum indicators: Compared with the blank group,the BNP of the model group increased,while ACE2,Ang(1-7),and MAS decreased,the difference was statistically significant(p<0.05).Shows that heart failure can reduce the expression of ACE2-Ang(1-7)-Mas.Compared with the model group,BNP decreased in the traditional Chinese medicine group and the positive drug group,while ACE2,Ang(1-7),and MAS decreased.The difference was statistically significant(p<0.05).It shows that Jiawei Baoyuan Decoction and Valsartan treatment can up-regulate the expression of ACE2-Ang(1-7)-Mas and improve the degree of heart failure,and there is no significant difference between the two effects(p>0.05).(3)mi R-1: Compared with the blank group,the mi R-1 of the model group was reduced,and the difference was statistically significant(p<0.05).Shows that heart failure can reduce the expression of mi R-1.Compared with the model group,mi R-1 increased in the traditional Chinese medicine group and the positive medicine group,and the difference was statistically significant(p<0.05).It shows that both Jiawei Baoyuan Decoction and valsartan treatment can up-regulate the expression of mi R-1,and there is no significant difference between the two effects(p>0.05).(4)MASSON staining: The cells in the normal group have normal morphology,and there are a few collagen fibers around them in uniform size.The morphology of myocardial cells in the model group is disordered,and there are a large number of collagen fibers around it,suggesting myocardial fibrosis.Fibrosis in the traditional Chinese medicine group and the positive control group was lower than that in the model group,indicating that valsartan and traditional Chinese medicine treatment can reduce the level of fibrosis.Conclusion: Jiawei Baoyuan Decoction can improve heart function and inhibit ventricular remodeling,and its mechanism may be related to the up-regulation of ACE2-Ang(1-7)-Mas and mi R-1.
