Objective: Observe the therapeutic effect of Linggui Yangxin Decoction on cardiac function in rats with chronic heart failure,as well as serum vascular terminal natriuretic peptide(NT-pro BNP),renin(PRA),angiotensin II(Ang?),aldosterone(ALD)vascular tone(1-7)[Ang(1-7)],Mas receptor(MasR)changes,analysis of the effect of Linggui Yangxin Decoction on RAAS and ACE2 / Ang(1-7)/ MasR axis of chronic heart failure rats,To explore its mechanism of treatment for chronic heart failure.Methods: 76 male healthy SPF grade SD rats were randomly divided into blank group,blank control group,model control group,Linggui Yangxin high,medium and low dose group(abbreviation: Linggui Yangxin H,M,L group),Bena In the Puli group,the blank and the blank control group had 8 animals in each group,and the remaining groups had 12 animals in each group.The blank group was fed normally,and the blank control group was intraperitoneally injected with an equal volume of normal saline.The remaining groups of animals were intraperitoneally injected with adriamycin(1.5mg / kg,once per week for 4 weeks)to establish a heart failure model.The blank group,blank control group and model control group were gavaged with distilled water,and the rest were gavaged with corresponding drugs for 4 weeks.During the experiment,observe the general situation of rats.After the administration,the levels of serum PRA,Ang?,ALD,Ang1-7,NT-pro BNP were measured by enzyme-linked immunosorbent assay(Elisa).Western blot was used to detect the protein expression level of MasR in cardiomyocytes,and the reverse transcription polymerase chain reaction(PT-PCR)method was used to determine the expression level of MasRmRNA in cardiomyocytes.Results:1.NT-pro BNP : Compared with the model control group,Linggui Yangxin H group,Linggui Yangxin M group,and Benazepril group decreased,the difference was statistically significant(P <0.01),Linggui Yangxin L The group was elevated,and the difference was not statistically significant(P> 0.05).2.PRA,Ang?,ALD: Compared with the model control group,the serum PRA,Ang?,ALD of Linggui Yangxin Group H,Linggui Yangxin Group M,Linggui Yangxin Group L,Benazepril group were significantly reduced.The difference wasstatistically significant(P <0.05).3.Ang1-7: Compared with the model control group,Linggui Yangxin Group H increased,the difference was statistically significant(P <0.05),Linggui Yangxin Group M,Linggui Yangxin Group L,Benazepril Both groups were elevated,and the difference was statistically significant(P <0.01).4.MasR protein and mRNA : Compared with the model control group,Linggui Yangxin group H increased,the difference was statistically significant(P <0.05),Linggui Yangxin group M,Linggui Yangxin group L,Bena Both in the Puli group were elevated,and the difference was statistically significant(P <0.01).Conclusion:1.Linggui Yangxin Decoction can effectively reduce serum NT-pro BNP levels and improve heart function in rats with heart failure;2.Linggui Yangxin Decoction can reduce serum PRA,Ang?,ALD levels,increase Ang1-7,MasR levels in rats with heart failure,may regulate the level of neuroendocrine by inhibiting the activity of RAAS,and increase ACE2 / Ang(1-7)/MasR axis exerts myocardial protection and is an effective prescription for treating chronic heart failure.
