Objective:To observe the changes of BWT score,cerebral infarction volume and cerebral D-Ser/NR1/Ca2+expression at different time points of electroacupuncture at pericardial meridian acupoints in MCAO rats,and to explore the regulation effect of electroacupuncture at pericardial meridian acupoints on neurotoxicity after cerebral ischemia,so as to provide some experimental basis for the scientific connotation of cardio-brain related theories.Methods:A total of 360 SD rats were randomly divided into normal group(120)and model group(240)for the first time.The left common carotid artery was inserted into the MCAO rat model.After successful modeling,240 model rats were randomly divided into model group and electroacupuncture pericardial meridian acupoint group for the second time,with 120 rats in each group.In each group,they were randomly divided into 5 time phase points groups at 1d,3d,7d,14d and 21d,with 24 rats at the time phase points.Normal groups are only bound,24h after modeling,the pericardium of the electroacupuncture group was treated with electroacupuncture.After binding,the rats were injected with no.15mm and no.36 filiform needles into the paralyzed side acupuncture point of the rats,with a depth of 3-4mm.The pericardium was divided into one group by tianquan and quze acupoints,and the other group by neiguan and daling,which the electroacupuncture instrument was connected.While the model group was bundled.for 30 minutes each time,once a day.BWT was used to evaluate the neurological deficits of rats before and after treatment.After treatment,the cerebral infarction volume,D-Ser content,NR1 expression and Ca2+content in of the rats in each group were detected by TTC staining,high performance liquid chromatography(HPLC),Western Blotting(WB)and Flow cytometry respectively.Result:1.Comparison of BWT scores in rats:After cerebral ischemia,the BWT scores of the model group and the pericardial meridian acupoint group were lower than those of the normal group(p<0.05).With the prolongation of ischemia time,the BWT scores of the model group at 14d and 21d and the pericardial meridian acupoint group at 3d,7d,14d and 21d were gradually higher than before(p<0.05).The scores of electroacupuncture pericardial meridians and acupoints group were increased at 3d,7d,14d and 21d after treatment(p<0.05).2.Cerebral infarct volume:After cerebral ischemia,the volume of cerebral infarction in the model group and the electroacupuncture pericardial meridian points group increased compared with the normal group(p<0.05).Compared with the model group at the same time point,the volume of cerebral infarction in the electroacupuncture pericardial meridian acupoints group was reduced at 7d,14d and 21d(p<0.05),and there was no significant difference at other time points(p>0.05).3.The content of D-Ser:After cerebral ischemia,compared with the normal group,the content of D-Ser in brain tissue of rats in each phase point model group and electroacupuncture pericardial meridian acupoint group increased(p<0.05).Compared with the same phase point model group,the content of D-Ser in electroacupuncture pericardial meridian acupoint group decreased on 3d,14d and 21d(p<0.05).4.The expression of NR1:After cerebral ischemia,the expression of NR1 in the brain tissues of the model group and the electroacupuncture pericardial meridian acupoint group was increased at all time points except for the electroacupuncture pericardial meridian acupoint group for 3d(p<0.05).The expression of NR1 in electro-acupuncture pericardial acupoints group was lower than that in the model group at the same time point(p<0.05).5.The content of Ca2+:After cerebral ischemia,except for the model group for 14d,the intracellular Ca2+content of the model group and the electroacupuncture pericardial acupoints group increased at each time point(p<0.05).Compared with the model group at the same time point,the content of the electroacupuncture pericardial meridians and acupoints decreased at 3d(p<0.05),and increased at 14d and 21d(p<0.05).Conclusion:1.Electroacupuncture at pericardial meridians can improve the cerebral infarction volume and neurological deficit in MCAO rats,and the improvement of neurological deficit is more obvious with the increase of treatment time2.The changes of D-Ser/NR1/Ca2+after cerebral ischemia show a certain pattern.Electroacupuncture at pericardium meridian can regulate the expression level of D-Ser/NR1/Ca2+,stably reduce the content of D-Ser,regulate the interaction between D-Ser/NR1,and regulate the expression of NR1 and Ca2+differently over time.3.Electroacupuncture at pericardial meridians may be involved in regulating excitatory neurotoxicity by regulating the expression of D-Ser/NR1/Ca2+,and promote the recovery of neurological function in MCAO rats.
