| Objective:To investigate the effect of Sangbaipipa paste on the mouse model of acne vulgaris induced by Propionibacterium acnes,to investigate the effects of Sangbaipipa paste on the naked eye observation,pathological section,oxidative damage level and inflammatory factor level of mice with acne vulgaris,and to explore the mechanism of Sangbaipipa paste in treating Acne vulgaris,and provides the experimental basis for the clinical application.Methods:ICR mice were used as experimental subjects,Sangbaipipa paste was used as intervention factor,tetracycline was used as positive control,Propionibacterium acnes was used to induce the auricle of mice,and the oxidative damage indexes were ROS,MDA,SOD,GSH,T-AOC,GSH,the levels of IL-1β and TNF-α were used as the index of effect in vivo.ICR mice were divided into 6 groups:PBS control group,model group,low-dose treatment group,middle-dose treatment group,high-dose treatment group and positive control group(tetracycline).PBS control group and model group were given 0.2 ml normal saline daily,the low,middle and high dose treatment group was given 0.2 ml Sangbaipipa paste solution per day,while the positive control group was given0.2ml diluted tetracycline suspension per day.After 14 days of treatment,the auricle of mice was photographed,the skin lesions of the auricle of mice were taken for pathological section,and the animals were killed after blood collection.Results:After 14 days of treatment,compared with PBS control group,the levels of ROS and MDA in model group were significantly higher(p<0.01),SOD、T-AOC and GSH were significantly lower(p<0.01).Compared with the model group,the levels of ROS and MDA in the treatment group were significantly lower than those in the control group(p<0.01),and the levels of SOD,T-AOC and GSH were significantly higher than those in the control group(p<0.01).Compared with PBS control group,the levels of IL-1β and TNF-α in model group were significantly higher(p<0.01).Compared with the model group,the levels of IL-1β and TNF-α in the treatment group were significantly lower than those in the control group(p<0.01).Conclusion:1.Propionibacterium acnes can induce oxidative damage and release of inflammatory factors.The synergistic action of the two factors has an effect on the course of acne.2.Sangbaipipa paste inhibits acne development by inhibiting oxidative damage,improving antioxidant capacity,and reducing levels of the body’s inflammatory cytokines.3.The function of Sangbaipipa paste to inhibit oxidative damage and reduce the level of inflammatory factors increased gradually with the increase of dosage,showing a good dose effect level.Objective:To investigate the effect of Sangbaipipa paste treated mice serum on HaCAT.To explore the mechanism of intervention of Sangbaipipa paste on the course of acne.Methods:HaCaT cells were divided into 5 groups:Blank Control Group,model group,low-dose group,middle-dose group,high-dose group,positive control group(Dexamethasone).In the first part of the experiment,the serum of "PBS control group","Model group"and"High-dose treatment group"mice was collected.Blank control group:HaCaT+10%control group;Model group:HaCaT+10%model group;Low dose group:HaCaT+2.5%high dose group mouse serum+7.5%FBS;Medium dose group:HaCaT+5%high dose group mouse serum+5%FBS;High dose group:HaCaT+10%high dose group mouse serum;positive control group(dexamethasone);H AC AT+10%dexamethasone mouse serum.The expression of RNA in IL-1β and TNF-α was detected by qPCR.The contents of ROS,MDA,SOD,T-AOC and GSH were detected by ELISA kit.Western blot was used to detect the expression of TLR-2,NF-κB,IL-1β and TNF-α.Results:① Compared with the control group,the protein contents of TLR-2 and NF-κB in the model group were significantly increased(p<0.01).Compared with the model group,the protein contents of Tlr-2 and NF-κB in middle-dose group,high-dose group and positive control group were significantly decreased(p<0.01);② Compared with the blank control group,the levels of IL-1β and TNF-α in the model group increased significantly(p<0.01),the mRN A and protein contents of IL-1β and TNF-α in low,middle and high dose group and positive control group were significantly decreased(p<0.01).③Compared with blank control group,ROS and MDA in model group were significantly increased(p<0.0 1),while SOD,T-AOC and GSH decreased significantly(p<0.01).Compared with model group,the levels of ROS and MDA in high dose group and positive control group were significantly lo wer(p<0.0 1),S OD,T-AOC and GSH were significantly higher(p<0.0 1).Conclusion:①S angbaipipa paste can decrease ROS content,decrease MDA activity and increase SOD,T-AOC,GSH activity,thus improve oxidative damage level.②Sangbaipipa paste exerts anti inflammatory effects by affecting the RNA and protein levels of inflammatory cytokines IL-1β and TNF-α.③Sangbaipipa paste may inhibit the expression of TLR-2 and NF-κB,inhibit the TLR-2/NF-κB signaling pathway,and down-regulate the level of IL-1β and TNF-α,thus improve the inflammatory response. |
