Objective: In this study,we used a post-infection cough(PIC)mouse model induced by endotoxin and stimulated by smoke and capsaicin to construct airway inflammation.By measuring cough frequency,Creactive protein(CRP)content in serum,expression of interleukin-1?(IL-1?)and Protein gene product 9.5(PGP9.5)in lung tissue of mice,To explore the effect mechanism of Shufeng Zhike Mixture on airway reactivity and inflammation in mice with post-infection cough.Methods:(1)Forty healthy male C57BL/6 mice were selected and divided into blank group,model group(PIC group),Shufeng Zhike mixture group(Chinese medicine group)and positive control group,with 10 mice in each group.The model was created by "smoke stimulation + endotoxin and capsaicin nose drops".(2)The drug was administered three times a day for a total of 5 days from the next day after successful modeling.The blank group and model group were given normal saline 7.5 m L /kg by gavage,the traditional Chinese medicine group was given Shufeng Zhike Mixture 2.76g/kg by gavage,and the positive control group was given compound methoxamine capsule 13.97mg/kg by gavage.(3)The general situation of mice was observed.After 5 days of intragastric administration,mice in each group were stimulated with ammonia to cough,and the cough times of mice were counted manually.Serum CRP content was detected by ELISA.The bronchial lung tissues were taken for paraffin section and HE staining.The changes of the tissues were observed under light microscope.Results:(1)The blank group was in good mental state,active and active,with smooth and shiny fur and normal diet;After modeling,the model group had poor mental state,less activity,less irritability,less hair gloss and less diet than the blank group.Shufeng Zhike Mixture group was similar to the positive control group,compared with the model group,the spirit was general,easy to irritate,and the diet was slightly reduced.(2)After 5 days of intragastoral administration of traditional Chinese medicine,mice in each group were stimulated with ammonia water.Data were analyzed by statistical software,and the normal distribution and homogeneity of variance were satisfied.One-way analysis of variance(F=16.29,P<0.05),the cough frequency of model group was increased compared with blank group,and the difference was statistically significant(P<0.05);Compared with model group,cough frequency of TCM group and positive control group was decreased,and the difference was statistically significant(P<0.05);Compared with the blank group,the cough of mice in Shufeng Zhike mixture group and positive control group was slightly increased,but there was no statistical significance(P<0.05).It is suggested that Shufeng Zhike Mixture can effectively reduce airway sensitivity and relieve the cough frequency of mice after infection.(3)The data of serum CRP content in mice satisfied the normal distribution and homogeneity of variance,and the analysis of variance of single factor(F=3.68,P<0.05).Compared with blank group,serum CRP content in model group was increased,and the difference was statistically significant(P<0.05),airway inflammation was present in mice;Compared with model group,serum CRP content in Shufeng zhike mixture group decreased,and the difference was statistically significant(P<0.05).The results showed that Shufeng Zhike Mixture could reduce the CRP content in serum and airway inflammation of infected cough mice.(4)By comparing the average optical density values of the positive products,the data were analyzed by statistical software,and the normal distribution and homogeneity of variance were satisfied.One-way analysis of variance(IL-1?: F=4.283,P<0.05;PGP9.5: F=22.15,P<0.05),model group,Shufeng Zhike Mixture group and positive control group had positive expression.Compared with the blank group,the degree of positive expression in the model group was increased,and the difference was statistically significant(P<0.05);Compared with model group,the expression of Shufeng Zhike Mixture group and positive control group was down-regulated,and the difference was statistically significant(P<0.05).The results indicated that Shufeng Zhike Mixture could reduce the expression of IL-1? and PGP-9.5 in lung tissue,and the decrease of IL-1? indicated that Shufeng Zhike Mixture could alleviate the hyperresponsiveness of airway.Conclusion: Shufeng Zhike Mixture can effectively reduce the CRP content in serum of mice with cough after infection,alleviate the inflammatory response of respiratory tract,down-regulate the expression of IL-1? and PGP9.5 in lung tissue,improve the neurogenic inflammation caused by the neuropeptide produced at the end of respiratory tract fibers,and reduce the sensitivity of airway reactivity and cough reflex.It improved the exposure of respiratory nerve fibers due to the damage of epithelial cells caused by infectious agents,and the abnormal distribution of nerve cells,thus intervening the development of PIC and reducing The Times of cough after infection.
