This study includes two parts.The method of modern fungal taxonomy,morphology combined with molecular biology techniques were used in the first part.The Hypoderma in Anhui Province were studied from the aspects of morphological anatomy,phylogenetics,geographical distribution and ecological environment.In the second part,molecular biology techniques were used to verify the new species based on the ITS r DNA sequence.The phylogenetic tree was constructed with ITS+n LSU and ITS+mt SSU+n LSU combined sequences to explore the phylogenetic relationship between genera and species of Hypoderma and its related groups.The specimens of Hypoderma collected from Yaoluoping National Nature Reserve and Huangshan Scenic Spot in Anhui Province,as well as the specimens preserved in the Forest Fungi Herbarium of Anhui Agricultural University(AAUF)were examined.A total of 12 species were identified,including 5 new species.They are Hypoderma circinatus,H.dendropanacis,H.kerriae,H.tiantangzhaiensis and H.yuexiensis.The results of phylogenetic tree based on double and multi-gene sequences showed that:Hypoderma,Lophodermium and Coccomyces were multi-originated,and Terriera was mono-originated.The geographical location of the host has a certain influence on the genetic relationship between the species of Hypoderma.The size and shape characteristics of ascospores play a certain role in the species identification of Hypoderma.The structural characteristics of the opening of the stroma play an important role in the division of the genus of Rhytismataceae.This study has important scientific significance in both fungi taxonomy and molecular phylogeny.
