With the rapid improvement of China’s economic living standard,people’s awareness of health care has been continuously improved,dual-purpose substances for food and medicine have been widely used,and the safety of their quality has aroused great concern of society.Some studies have shown that some dual-purpose food and medicine substances will produce some toxic and side effects when eaten for a long time or in large quantities.If Polygonum multiflorum is used for a long time,it will damage the liver,and aristolochic acid compounds are toxic to the kidney.Therefore,it is of great practical significance to strengthen the monitoring of some endogenous components in dual-purpose food and medicine substances to ensure people’s health and promote the sustainable development of social economy.In this thesis,some endogenous components in Houttuynia cordata,Prunella vulgaris L.and Gardenia jasminoides were selected as the research objects,and the new analytical methods of endogenous components in the above-mentioned edible and medicinal substances was established by using liquid chromatography and liquid chromatography-mass spectrometry analytical techniques.It was applied to the analysis of samples from different places in China,and achieved good results,which provided a certain theoretical basis for the safety risk assessment of the above-mentioned dual-purpose food and medicine substances in the future and technical support for the effective monitoring of their quality.The main research results and conclusions are as follows:1.A rapid and simple analytical method was established for the simultaneous determination of analogues of aristolochic acids(aristolochic acid I,aristolochic acid II,aristolactam I and aristolactam AII)in Houttuynia cordata by ultra-high performance liquid chromatography–quadrupole/time-of-flight mass spectrometry(UHPLC–Q/TOF-MS).The samples were ultrasonically extracted with methanol,purified on HC-C18.The extraction and purification conditions were optimized in detail.Under the optimized conditions,aristolochic acid I(0.08-2 mg/L),aristolochic acid II(0.04-2 mg/L),aristolactam I(0.04-2 mg/L)and aristolactam AII(0.004-2 mg/L)all showed good linear regression(R~2>0.9987),LOD and LOQ were in the range of0.0001-0.02 mg/L and 0.0003-0.05 mg/L mg/L,respectively.The average recoveries of the four analytes were in the range of 72.3%–105.5%,with the relative standard deviations(RSDs)≤7.6%.The proposed method was then applied to the determination of aristolochic acid I,aristolochic acid II,aristolactam I and aristolactam AII in Houttuynia cordata samples collected from 22 different regions of China.The content of aristolactam AII was between 0.67 mg/kg and 17.24 mg/kg,and the other three target compounds were not detected.2.A new analytical method was established for simultaneous determination of rosmarinic acid and ursolic acid in Prunella vulgaris L.from different regions using ultra-high performance liquid chromatography.The samples were ultrasonic extracted with 50%ethanol,and the extraction conditions were optimized,primary secondary amine(PSA)was used to purify the extract.Under the optimized conditions,rosmarinic acid(2-200 mg/L)and ursolic acid(1-100 mg/L)showed good linear regression in the selected concentration range(R~2>0.9994).The detection limits(LODs)were 0.02 mg/L and 0.14 mg/L,and the quantification limits(LOQs)were 0.05 mg/L and 0.48 mg/L,respectively.Under the optimized conditions,the average recoveries of the two analytes were within the range of 80.5-119.6%with the relative standard deviations(RSDs)≤2.6%.The proposed method has been well applied to the determination of Prunella vulgaris L.samples collected from 9 different regions in China.The contents of rosmarinic acid and ursolic acid were 3.97-20.01 mg/g and 1.53-5.86 mg/g,respectively.3.A new analytical method was established for the simultaneous determination of iridoids(geniposide,genipin and geniposidic acid)in Gardenia jasminoides by ultra-high performance liquid chromatography-photo diode array(UHPLC-PDA).The samples were ultrasonic extracted with water and the extraction conditions were optimized.Under the optimized conditions,geniposide(0.3-300 mg/L),genipin(0.1-50 mg/L)and geniposidic acid(0.4-200 mg/L)all showed good linear regression(R~2>0.9984),LOD and LOQ were in the range of 0.03-0.11 mg/L and 0.10-0.38 mg/L mg/L,respectively.The average recoveries of the three analytes were in the range of104.5%-119.7%,and the relative standard deviations(RSDs)≤3.7%.The proposed method was then applied to the actual sample analysis.
