Analysis Of Alkaloid Component And Toxicity Test In The Pedicularis Cheilanthifolia

Abstract/Summary:
Taking Pedicularis cheilanthifolia collecting from Bayinbuluke,Xinjiang on as the object of the research,this study detected the types and amounts of alkaloids of P.cheilanthifolia by TLC and GC-MS,and determined the LD50by acute toxicity test in mice and observed the changes of blood physiological and biochemical indexes,histopathology,and other aspects of subchronic toxicity in mice.it was provided theoretical evidence for toxicology,mechanism of toxicity,utilization prospect of P.cheilanthifolia.In this study,following results were obtained.The results are as follows:1.Analysis on alkaloids:Alkaloids contained in P.cheilanthifolia were extracted,separated and detected using alkaloid extraction,TLC,column chromatography and GC-MS.The results showed that the extract rates of acidity dichloromethane extraction section,alkaline dichloromethane extraction section and alkaline n-butanol extraction section were 2.0%,0.17%and 10.3%,respectively.The acidity dichloromethane extraction section has no alkaloids,the alkaline dichloromethane extraction section contains at least 5 alkaloids,the alkaline n-butanol extraction section contains at least 2 alkaloids.By column chromatography and GC-MS,acidity dichloromethane extraction sectiont does not contain alkaloids.There are24 kinds before column chromatography and 20 kinds after column chromatography on alkaline dichloromethane extraction section.Among all the alkaloids,6 kinds alkaline on alkaline dichloromethane extraction section before and after column chromatography are same.There are 8 kinds of alkaloids before column chromatography and 9 kinds of alkaloids after column chromatography on alkaline n-butanol extraction section.Among all the alkaloids,1 kind alkaline on alkaline n-butanol extraction section before and after column chromatography is same.Alkaloids were detected in P.cheilanthifolia,such as3-acetyllycopsamine,sophocarpine,O6-demethylmenisporphine,9-methoxyellipticine,etc.2.Acute toxicity test in mice:In order to determine the dose of LD50and evaluate the acute toxicity of P.cheilanthifolia,the mouse were given the water extraction solution of P.cheilanthifolia by irrigating.The test results showed that the LD50of gavage was 147.4 g/kg,and the 95%confidence limit was 129.5 to 167.6 g/kg.Combined with a number of indicators such as the degree of clinical symptoms of poisoning,pathological necropsy and incubation period of poisoning in the test mice,it is considered that there is slightly poisonous in P.cheilanthifolia.3.Subchronic toxicity test in mice:By feeding different doses(15%,30%)of P.cheilanthifolia for100 d,the clinical manifestation,feed intake,weight,organ coefficient were observed and serum physiological parameter,hematological parameters and histopathology were analyzed.The results showed that there was no significant change in the clinical symptoms of mice by feeding different doses of P.cheilanthifolia.The feed intake was significantly higher than that of the control group and there was no significant difference in body weight compared with the control group.During the experiment,the physiological index fluctuated,but it returned to normal at the end of the test.The CRE,GLU of mice in high dose group was significantly higher than the control group.The organ coefficient of liver in the high dose group was significantly increased compared with the control group,while the other differences were not significant.Pathological examination showed that it caused some damage to kidney and liver of mice and had slight effects on the heart,spleen,lungs and brains of mice and no effect on the cerebellum of mice by feeding 30%doses of P.cheilanthifolia.
