| Rapeseed(Brassica napus L.)is one of the most important oilcrops worldwide,which has significant heterosis.Significant heterosis for seed yield and other agronomic traits has been well documented in rapeseed.Safe,reliable and efficient pollination control system is the base for heterosis utilization.Because of its sterility stability,complete cytoplasmic resources,absence of negative cytoplasmic effects and so on,genic male sterility(GMS)has become the main pollination control system in the utilization of heterosis in rapeseed.Shaan-GMS was a natural mutant male sterile accession discovered in 1994 by Northwest Agriculture and Forestry University Rapeseed Research Center.Previous studies showed that male sterility of Shaan-GMS is controlled by a monogenically multiallelic locus with 3different alleles:Mf,Ms,ms.Ms is a sterile allele,Mf is a restorer allele,that suppresses the expression of Ms,and ms is a fertile allele.The relationship among them is Mf>Ms>ms(Chen2015).According to this hypothesis,homozygous and heterozygous two-type lines of Shaan-GMS are selected.In this research,the homozygous two type lines of Shaan-GMS803AB(Ms Ms×Mf Ms)was used as a material,and we exploited a set of markers based on AFLP technique and whole genome sequencing strategy,and constructed a genetic linkage map of Mf genes.The main results are as following:1.The male sterile plants and fertile plants in Shaan-GMS 803AB showed obviously difference for their floral organs.The flowers in fertile plants had 6 anthers,which released a huge of yellow pollens.The sterile plants flowered late,with stamens being severely degenerated,being shorted than pistil.The sterile buds were dry and slightly wrinkled.2.The population of Shaan-GMS 803AB contained 646 plants,of which 299 were fertile and 347 sterile,which fit a 1:1 segregation ratio(χ2=3.567).There were 479 plants in F2populations,of which 344 were fertile and 135 were sterile,which fit a 3:1 segregation ratio(χ2=2.589).3.The 5 molecular markers(IP5F2R2,IP60F2R2,IP65F2R2,IP74F2R2 and CAPS26080)developed previously were used to genotype 646 plants in the Shaan-GMS 803AB for constructing a genetic linkage map for the restorer gene Mf.The results showed that the closest markers on the two sides of the Mf gene were IP5F2R2 and CAPS26080,respectively.The genetic distance to the Mf gene was 0.6 c M and 0.9 c M,respectively.4.The same set of markers were also used to genetype 479 plants in a F2 population deriving from Shaan-GMS 803AB.As a result,16 recombinant individuals were found.Finally,a genetic linkage map was constructed with all of the five markers on the same side of the Mf gene.The genetic distances of markers IP5F2R2,CAPS26080,IP60F2R2,IP74F2R2and IP65F2R2 to the Mf gene were 0.2 c M,1.2 c M,1.6 c M,2.4 c M and 4.4 c M,respectively. |
