| Polygonatum cyrtonema Hua is a perennial herb of the genus polygonatum of the lily family.Its medicinal part is the rhizome.It is difficult for Polygonatum cyrtonema Hua seeds to germinate,this study used Polygonatum cyrtonema Hua seeds establish micropropagation system,screening suitable pretreatment method and germination medium,medium of induction of multiple shoot,and rooting medium and screening suitable transplanting matrixes,at the same time using sand and hormone treatment to break the seed dormancy,screening suitable pretreatment method and the seeding matrix,in order to shorten the germination time of Polygonatum cyrtonema Hua seeds,improve the germination rate,provide reference for breeding of Polygonatum cyrtonema Hua seedlings.The research results are as follows:1.through the four factors three levels orthogonal test,select Polygonatum cyrtonema Hua seeds germination rate higher processing method,namely 4℃ hidden 60 d,then 20℃,and 120 W ultrasonic processing 20 min,soaked in concentration of 180 mg/L,temperature of 40℃ of GA3 in 24 h,the last vaccination in MS+1.0 mg/L 6-BA+1.0 mg/L GA3+0.1 mg/L NAA medium.2.Through the orthogonal test of three factors and three levels,the appropriate combination of Polygonatum cyrtonema Hua seed tissue culture seedlings to induce cluster bud medium was selected,namely MS+2.0mg/L 6-BA+1.5mg/L GA3+0.3mg/L NAA.After screening different rooting media,it was concluded that the appropriate combination of rooting media was suitable for inducing Polygonatum cyrtonema Hua seedlings,namely MS+0.1mg/LIBA+activated carbon.After screening the survival rate of Polygonatum cyrtonema Hua tissue culture seedlings with different transplanting substrates,it was concluded that transplanting seedlings were easy to survive.In rural soil:sand=1:1,rural soil:sand:vermiculite=1:1:1,and rural soil:sand:vermiculite=5:1:1,the survival rate was the highest,which was 100%.3.Field experiments showed that the germination rate of seeds soaked by sand and hormones in the mixed soil and sand matrix was as high as 92%after 60 days of field germination,and the emergence rate of seeds in the mixed soil and vermiculite matrix was 82.35%after 90 days.4.On the basis of field production test,the technical rules of sand storage of Polygonatum cyrtonema Hua seeds in qingyang and suitable areas of anhui province were summarized. |
