| Lotus is a perennial emergent flower of the genus Nelumbo,which is widely distributed in the world.It is not only rich in Germplasm and elegant in appearance,but also has high ornamental and economic value.At present,there are few studies on the genetic diversity of lotus,but there are few reports on the population structure and association analysis of lotus.In this paper,72 lotus germplasm resources collected by Henan high quality flower and vegetable seedling Engineering Research Center were selected as the research objects.The phenotypic data of 10 quality characters and 17 quantitative characters were statistically analyzed.At the same time,16 pairs of primers were used to amplify SSR molecular markers to analyze the genetic diversity of Lotus germplasm resources.At the same time,the association analysis between phenotypic traits and SSR markers was carried out on this basis,and the main research results are as follows:(1)The genetic diversity index of 10 quality characters of 72 lotus materials ranged from 1.05 to 2.12;the coefficient of variation of 17 quantitative characters ranged from 20.91%to 63.22%,the average coefficient of variation was 32.34%,and the genetic diversity index ranged from 6.08 to 13.44,the average value was 6.56.The results of correlation analysis and principal component analysis showed that there were complex relationships among the traits,and the cumulative contribution rate of the first eight principal components was 77.778%.The D value of No.4 is the highest,which indicates that its comprehensive performance is relatively the best.There are six groups in Euclidean distance 6.0.(2)A total of 96 alleles were amplified by SSR markers,and the average number of alleles was 6;the average value of Shannon diversity index was 1.170 7,and the average value of polymorphism information content was 0.524 2.The genetic similarity coefficient of 72 lotus materials ranged from 0.386 to 0.912,with an average of 0.670.Based on UPGMA method,the cluster graph can be divided into 8 groups at the similarity coefficient of 0.67.According to the analysis of population genetic structure,lotus materials were divided into three groups.(3)The phenotypic traits were associated with SSR markers revealed that 13 SSR loci are associated with 12 phenotypic traits.The interpretation rate of trait variation is between 15.44%-74.55%,with an average of 33.01%,and the interpretation rate among traits The gap is relatively large,with the highest anther length(74.55%)and the lowest flower status(15.44%).The above results showed that the genetic diversity of 72 lotus materials was rich,which provided a scientific basis for the identification and evaluation of Lotus Varieties and the breeding of new varieties... |
