Pinus taiwanensis Hayata is an endemic tree species in China.It is an important tree species for greening,afforestation and ecological restoration in the subtropical high-altitude mountains of eastern my country.The ecological,social and economic value of Pinus taiwanensis is one of the hosts of pine wood nematode.With the continuous spread of pine wood nematode,it poses a huge threat to Pinus taiwanensis resources in Huangshan Mountain.However,there have been no reports on the research on the conservation and tissue culture and rapid propagation system of Pinus taiwanensis germplasm resources.In this study,Pinus taiwanensis seed bulbs at different altitudes were used as experimental materials,and the seeds obtained from the seed bulbs were stored at different temperature gradients to study the effects of different storage conditions on seed vigor.At the same time,the embryo of Pinus taiwanensis seeds was used as the experimental material to construct the Pinus taiwanensis tissue culture and rapid propagation system.The specific results are as follows:The seeds of Pinus taiwanensis at different altitudes(1200 m,1400 m,1600 m,1800m)were stored in 3 temperature gradients(4℃,-20℃,-80℃),and the germination rate and germination rate of the seeds were checked regularly.At the same time,the tissue culture and rapid propagation system were studied with the embryos of the stored Pinus taiwanensis seeds.The experimental results showed that:1.The study on the effect of seed storage conditions on seed vigor shows that the vigor of seeds stored under low temperature(-20℃,-80℃)conditions is significantly higher than that of 4℃ storage conditions.2.The tissue culture and rapid propagation system of Pinus taiwanensis has been established,and the specific cultivation conditions are as follows:1)The best disinfection combination of Pinus taiwanensis seed embryos is 75% alcohol disinfection for 1 minute and 3% sodium hypochlorite for 10 minutes;2)The best induction medium for seed embryo callus is MS+1mg/L 6-BA+0.2 mg/L NAA;3)The best induction medium for adventitious buds is DCR+1 mg/L 6-BA+0.05 mg/L NAA;4)The best induction condition for adventitious bud elongation is DCR+0.1 mg/L 6-BA+0.05 mg/L NAA;5)The best medium for rooting effect is 1/2 DCR+2 mg/L IBA+0.05 mg/L NAA.By carrying out research on the germplasm resources of Pinus taiwanensis,the preliminary establishment of the rapid propagation system of Pinus taiwanensis will be of great significance for the preservation of Pinus taiwanensis germplasm resources,genetic improvement,cultivation of new varieties(lines),and reproduction of high-quality seedlings.
