System of tissue culture and plant regeneration is essential for genetic transformation. In Buffallograss tissue culture, seedling segment, leaf base, mature embryo and immature male inflorescences can be used as explant. Calluses of Buffallograss was inducted mostly on MS medium with 2.0mg/L 2,4-D+ 10.0mg/L AgNO3; the subculture and shoot regeneration was achieved on MS+6-BA applie with other nutriments. And the medium of root production was 1/2 MS+6-BA or MS+NAA. Rooted plantlets could be successful transferred to a soil mixture with a ratio of 1 soil : 3 vermiculite : 3 perlite : 4 sphagnum moss peat.The study on Buffallograss tissue culture and regeneration systems was conducted though male immature inflorescences culture and seed in the article.1. Mature embryos culture system: Buffallograss seeds were inoculated on MS medium with 2.0mg/L 2,4-D, 3% agar, pH 5.8 for callus induction for 20 days in dark. The embryo calluses obtained form seed was transferred on B5+2.0mg/L 2,4-D+1.5mg/L 6-BA + 50mg/L CH for shoot regeneration. Subculture on MS + 2.0mg/L 6-BA + 50.0 CH medium could improve shoot regeneration. The medium of root production was on 1/2MS +0.5mg/L NAA or 1/2MS +1.0 NAA for 24h/d illumination period.2. Male immature inflorescences explant system: Calluses was inducted on medium MS +2.0mg/L 2,4-D + AgNO3 +3% agar, pH 5.8 from male immature inflorescences for 20 days in dark.. Both subculture and shoot regeneration of embryo calluses from induction medium was conducted on MS+2.0mg/L 2, 4-D+0.1mg/L 6-BA. The root regeneration could be succeeded on medium 1/2 MS +0.3mg/L6-BA.
