Walnut is widely distributed in China,and its fruit is rich in nutritional value and its wood is hard.Walnut is an important woody grain and oil species.Somatic embryogenesis of walnut plays an important role in rapid breeding and molecular breeding.Previous studies showed that walnut regeneration differed greatly with different genetic bases.’Xilin no.3’ walnut potential exuberant,adaptability,high yield strong.At present,there are few studies on somatic embryogenesis of ’Xilin 3’walnut.In this study,’Xilin 3’ walnut was used as experimental material to explore the influencing factors of somatic embryogenesis and maturation of ’Xilin 3’ walnut,and the regeneration system of somatic embryogenesis of ’Xilin 3’ walnut was preliminarily established,which laid a foundation for genetic transformation of ’Xilin3’ walnut.The main research results are as follows:1.In the disinfection treatment of explants,the best method was to use 70%alcohol to sterilize seeds for 30 s,and the survival rate was about 61% at the 4th day of inoculation.The best disinfection method for young embryos was 70% alcohol for5 min,and the survival rate was about 83% on the 4th day.2.The embryogenic callus induction rate of explants was significantly different at different sampling periods.In the experiment,the embryogenic callus induction rate of young embryos and cotyledons around May 31 was about 85%,and the embryogenic callus induction rate of seeds around April 30 was about 25%.3.The embryogenic callus induction rate reached 38% when the modified DKW was supplemented with 1.0mg/ l 6-ba +2.0mg/LKT+0.01mg/ l LIBA as explants.4.In the process of somatic embryogenesis,the modified DKW medium supplemented with 0.5mg/ l 6-ba +1.0mg/LKT+0.005mg/ l LIBA was the optimal medium for callus proliferation,and the callus proliferation rate was about 97%,and somatic embryos were formed.5.’Xilin 3’ walnut somatic embryos placed in the dark culture for 2 weeks and then transferred to light culture for 2 weeks of light treatment can make the normal development of somatic embryos,15% walnut somatic embryos into normal plants.
