Paeonia decomposita belongs to the Paeonia peony group of the Paeoniaceae.It is one of the nine wild peony species unique to China.As a precious wild peony resource,Paeonia decomposita is the second batch of planned plants for key protected wild plants in China.Paeonia decomposita not only has ornamental and medicinal value,but its peony seeds also have great potential in oil development.In recent years,Paeonia decomposita has been endangered due to man-made activities and severe growth environment.The current research reports on Paeonia decomposita mainly focus on taxonomy,microbiology,physiology and biochemistry,etc.,and there is a lack of a system based on the chemical substance of Paeonia decomposita.Research is not conducive to the utilization and protection of this species.This study uses a variety of analytical methods to determine the organic components and inorganic elements of Paeonia decomposita,combined with traditional Chinese medicine fingerprinting technology to explore the common characteristics and differences of the chemical components of Paeonia decomposita in different growing areas,and provide methods and basis for understanding the chemical composition characteristics of Paeonia decomposita,Which aims to provide scientific basis and basic data for the effective protection of this resource,introduction and cultivation,wild domestication,and selection and breeding of oil peony.The specific research content and results are as follows:Paeonia decomposita plants and rhizosphere soil samples were collected from both banks of the Dadu River Basin,specifically Kangding City and Danba County in Ganzi Prefecture,and Xiaojin County,Li County,Jinchuan County,and Malkang City in Aba Prefecture.About 20 samples of the roots,seeds,stems,leaves and rhizosphere soil of Paeonia decomposita were collected,and the roots were stripped into two parts,phloem and plug.Gas chromatography-tandem mass spectrometry(GC-MS)and ultra-high performance liquid chromatography(UPLC)were used to determine the organic components in Paeonia decomposita phloem.According to the results of GC-MS,54volatile components in 17 Paeonia decomposita cortex samples were preliminarily deduced,including 29 components in total.Phenolic glycosides and fatty acid compounds account for more than 80%of the total amount of methanol extracted compounds,of which paeonol has the highest content,with a relative content of about50%.According to the analysis results of principal component analysis and orthogonal partial least squares analysis(OPLS-DA),12 compounds with VIP values greater than1 that caused differences in the volatile compound content of phloem from different growing areas were screened out.These 12 characteristic compounds reflect The difference of volatile components of phloem in different growing areas.Established a method for analyzing the UPLC fingerprint of Paeonia decomposita phloem and a quantitative analysis method for the determination of four active ingredients.Combined with the Chinese medicine chromatographic fingerprint similarity evaluation system software(2012 version),17 Paeonia decomposita phloem fingerprints and standard fingerprints were established,and the similarity of the samples was evaluated.The results showed that the established UPLC fingerprints could reflect the Paeonia decomposita phloem fingerprints.The organic composition characteristics of the skin have good reliability and effectiveness.The contents of 4active ingredients(paeonol,paeoniflorin,paeonol protoside and benzoylpaeoniflorin)of 17 phloem samples were determined at 7.0~24 mg·g-1、5~37 mg·g-1、4.1~74 mg·g-1 and 4.8~20 mg·g-1.Analyzed and compared the fingerprints and active ingredient content of Paeonia decomposita phloem and two medicinal peony produced in Anhui,and compared with the effective ingredient content of medicinal peony in many provinces in the literature.The results showed that Paeonia decomposita The content of effective medicinal ingredients in phloem is higher than that of other medicinal cortex,and it has high medicinal value.Finally,using SPSS elements to cluster the common peaks of Paeonia decomposita phloem in different growing areas,the results show that there is no obvious growth area clustering law,indicating that the content of effective components in the phloem has nothing to do with the administrative division of the growing area but with the specific growth environment related.The Paeonia decomposita plants(phloem,plug,stem,leaf,seed)were determined by high-pressure airtight microwave digestion method,inductively coupled plasma mass spectrometry(ICP-MS)and inductively coupled plasma atomic emission spectrometry(ICP-OES).And the content of 18 inorganic elements in rhizosphere soil.The content and distribution characteristics of inorganic elements in plants were discussed,and a basic understanding of inorganic elements in Paeonia decomposita was obtained.The characteristics of the accumulation and migration of Paeonia decomposita plants to the inorganic elements in the rhizosphere soil were discussed.The results showed that the phloem of Paeonia decomposita in different growing areas has basically the same enrichment capacity for inorganic elements,and there are more elements with a migration coefficient greater than 1 in the leaves than in the seeds.It shows that inorganic elements are easier to migrate and accumulate in leaves.The correlation between Paeonia decomposita bast and inorganic elements in seeds,the correlation with the inorganic elements in rhizosphere soil,the correlation between the elements in bast and the four active ingredients,and the results showed that in the phloem In the seed samples,there is a significant positive correlation or a significant negative correlation between multiple pairs of elements,indicating that there is a synergistic or competitive relationship when these elements are absorbed by plants.The correlation between rhizosphere soil and inorganic elements in phloem and seeds has complex and diverse characteristics.There is a significant positive correlation or a significant negative correlation between the four active ingredients and different elements,indicating that the elements promote or inhibit the synthesis and accumulation of the active ingredients.Understanding Paeonia decomposita’s enrichment and migration characteristics of inorganic elements and the correlation of element content can provide a reference for subsequent introduction and cultivation,selection of planting bases,and fertilization.The fingerprints and standard fingerprints of 18 inorganic elements in Paeonia decomposita phloem and seeds were established using Origin software,and the similarity of sample fingerprints was evaluated by the vector similarity method.The results showed that most of the phloem sample elements were from different growing areas.The content and distribution law are highly similar,while the element content and distribution law of seed samples from different growing areas are relatively similar,and the content of most elements is not significant.The established element fingerprint can reflect the overall characteristics of inorganic elements in phloem and seeds.Principal component analysis and OPLS-DA analysis were carried out using R software and SIMCA software,and the characteristic inorganic elements that caused different growth of phloem and seeds were found.The characteristic inorganic elements in the phloem are B,Ti,Zn,Ba,Mo,Ca,Mg,Pb,the characteristic inorganic elements in seeds are B,Cu,Fe,K,and Zn.These characteristic elements reflect the differences of inorganic elements in bast and seeds in different growth areas.
