The genus of Chimonanthus Lindl. was only find in China, include Ch. praecox(L)Link.、Ch. Zhejiangensis M. C. Liu、Ch. salicifolius S. Y. Hu、Ch. nitens Oliv、Ch.grammatus M. C. Liu、Ch. campanulatus R.H.、Ch. campanulatus var. guizhouensis R. H.Chang et C.S.Ding var.guizhouensis R.H. Chang, Chimonanthus plant was beautiful andcould be used in medicine. Based on the investigation and collection of the aromaticcompounds species distribution on Chimonanthus, and through research of molecularmarkers and the main secondary metabolites, the results are as follows:To establish the HPLC fingerprints of genus Chimonanthus leaves, C18column (4.6mm×250mm,5μm) was used with the mobile phase containing acetonitrile-water. Forgradient elution, acetonitrile(B),0-20min,6%-20%;20-30min,20%-25%;30-40min,25%-45%;40-50min,45%-80%;50-80min,80%-85%;80-90min,85%-100%;90-110min,100%. Flow rate0.8mL·min-1and wavelength228nm. The temperature of columnwas25℃. The HPLC fingerprint of the chemical composition on the genusChimonanthus leaf was first constructed. The common peaks in the fingerprint spectra ofthe five reference substances of the scopoletin-7-O-β-D-glucoside, kaempferol-3-O-β-D-rutinoside, kaempferol-3-O-β-D-glucoside, quercetin and kaempferol were identified, andthe quantification was significant different. HPLC fingerprints of the Ch. Praecox, Ch.nitens, Ch.salicifolius, Ch. Zhejiangensis and Ch. grammatus were identified24,30,22,34and30common peaks, respectively. Different kinds and content of chemical compositionsin the different kinds of species were confirmed through the HPLC fingerprint.The contents of the five major chemical constituents on Chimonanthus wereinvestigated though the HPLC analysis. Quantify of the same compound in differentspecies was confirmed significantly different. On the development and utilization of thechemical compositions of the leaves of Chimonanthus should be paied attention to thespecies and the harvest time.The essential oil of Ch. salicifolius leaves were obtained by steam distillation. Thechemical components were separated and identified by gas-chromatography-massspectrometer (GC-MS). Sixty-five compounds were separated, of which sixty-onecompounds were identified. The essential oil components were rich in Ch. salicifolius.Essential substances are different in quantity and quality in different month. This method isgood for separating and analyzing the components of the essential oil of Ch. salicifoliu leaves, and can provide reference for studies on the exploitation and application of theessential oil of Ch. salicifolius. Plants in physiological activity produce more secondarymetabolites. By comparative analysis of five compound contents of Ch. salicifolius, thecompound annual dynamic law can be drawn and it can determine a reasonable harvestperiod.Using methods of headspace GC-MS to identify different provenance ofChimonanthus,27kinds of chemical compositions were detected in actual, especially inCh. grammatus. The method played an important role in quality control on Chimonanthusand can be widely used in the detection of other resources analysis and classification. Thehigh-quality provenance of Chimonanthus can be confirmed by comparative analysis offive compound contents in different provenance.In use of ISSR molecular marker technique, the genetic diversity of21populations ofthe aromatic leaf species in Chimonanthus was analyzed. The result showed that thearomatic leaf species in Chimonanthus were low in genetic diversity. The total geneticdiversity, observed number of alleles, effective number of alleles, Nei’s genetic diversityindex and Shannon’s information index were1.7160,1.3790,0.2220and0.3388,respectively. Therefore, the protection of species resources should be more valued. Formthe molecular level, the results in accordance with which aromatic leaf species ofChimonanthus were divided into Ch. salicifolius, Ch. Zhejiangensis, Ch. nitens and Ch.grammatus.
