| In this experiment,the fresh seeds of ’Moqiu’ cultivar in Aronia melanocarpa were selected as the test material,and the biological characteristics and the method of breaking seed dormancy,as well as the changes in the content of seeds stored under 90-day stratification were studied,and seed completion time to break dormancy after ripening;changes in related enzyme activities;changes in hormone content and analysis of seed proteomics.It provided a reference for exploring the seed dormancy mechanism and the metabolic process involved in seed protein of Aronia melanocarpa.The dormancy type of Aronia melanocarpa seeds is morphophysiological dormancy.1.The seeds of Aronia melanocarpa are kidney-shaped,with dense seed coats,and the thousand-seed weight is about(4.8±1.0)g.Water absorption of seeds increased with time,and reached the highest at 144 h.Under the treatment of 98% H2SO4,the germination rate of seeds was 0%.When the Na OH concentration was 1mol/L,the germination rate of seeds was higher than other treatments.When the seeds were soaked in hot water at 60℃,the germination rate of the seeds reached 67.3%,and the germination effect was better.And the seeds began to germinate after the post-ripening process on the 75 th day of stratification.The dormancy type of Aronia melanocarpa seeds is morphophysiological dormancy.2.During the 90-day stratification process,the content of soluble sugar and MDA increased first and then decreased,reaching the highest value of 3.75 mg/g and 0.0431 μmol/g on the 60 th day of stratification.Starch and soluble protein levels continue to decline.Starch and soluble protein were decomposed into easily absorbed small molecular substances,and the increase of soluble sugar could provide the energy needed for seed germination.Malondialdehyde can cause damage to cell membranes,and the decrease of its content indicates that the water vapor in the seeds permeates more easily,which is more conducive to the germination of seeds.3.During 90 days of stratification,the content of IAA in Aronia melanocarpa seeds increased first and then decreased,the content of GA3 is continuously rising,the content of ABA fluctuated,interaction between hormones to promote seed germination.In the process of stratification,the activities of SOD and G-6-PDH in the seeds of Aronia melanocarpa increased first and then decreased.The activities of POD increased,while the activities of CAT and ACP fluctuated.The reason for these phenomena is that the respiration is enhanced when the seeds begin to germinate at the later stage.These enzymes are related to the PPP cycle pathway,and the increased enzyme activity promotes the PPP pathway and accelerates seed germination.4.There are 43 differential proteins in Aronia melanocarpa seeds during stratification,of which 29 proteins are significantly up-regulated and 14 are down-regulated during the stratification process.The isoelectric points of the differential proteins are concentrated between p H 4.5 and 6.5,the molecular weight is mainly between 10 and 40 KDa.According to the GO classification criteria,the differential proteins in seeds mainly involved binding process,catalytic process and cellular process.Annotated with KEGG software,the results showed that the differential proteins were mainly involved in the ribosomal pathway,carbon metabolism pathway,RNA transport pathway,glycolysis pathway,protein processing pathway in the endoplasmic reticulum,amino acid biosynthesis pathway,and biosynthetic pathway of secondary metabolites. |
