Canine skin trauma is a common surgical disease,and the main routine treatment method used by veterinarians in clinical practice is instrument suture combined with drug therapy.As a popular herbal medicine in recent years,Ganoderma lucidum spore powder is mainly used in daily health care to improve immunity and anti-tumor.And based on its known pharmacological effects of immune modulation and antioxidants,it is applied to the treatment of canine skin wounds for the first time.This project aims to verify the feasibility of Ganoderma lucidum spore powder in the treatment of skin wounds and to explore its mechanism of action.In this experiment,10 Chinese pastoral dogs were used for wound treatment,and they were randomly divided into two groups.The experimental group was fed with prescription food for promoting wound healing,and the control group was fed with ordinary diet.Compare and validate the usability of the trauma model.The subject used 12 Chinese pastoral dogs.Five round wounds with a diameter of 15 mm were created on the back of the dogs with a surgical punch.The wounded dogs were randomly divided into 2 groups(n=6).The experimental group added Ganoderma lucidum to the daily diet.Spore powder(3.76 g/kg),the control group did not add Ganoderma lucidum spore powder,and the two groups were fed normally.At 0d,3d,7d,14 d,and 21 d,the wounds were photographed and recorded,the wound conditions were observed,the wound healing rate was calculated in each period,and the evaluation dogs’ mental status and wound healing time were recorded.At 7d,14 d,and 21 d,the skin wound tissue was collected by surgery.The sections of skin wounds were stained by HE and MASSON respectively to observe the deposition of inflammatory cells and collagen,and to record the micro vascularization and decline,the levels of malondialdehyde(MDA)and superoxide dismutase(SOD)in skin wounds were detected,detect the levels of prostaglandin E2(PGE2),tumor necrosis factor-alpha(TNF-α)and interleukin 1α(IL-1α)in skin wound tissue,detect the levels of Ig A,Ig M,and Ig G in skin wound,detect platelet-derived factor(PDGF),basic cell growth factor(b FGF)and vascular endothelial growth factor(VEGF)gene relative expression.Validation results of trauma model: There was no significant difference in healing between the wounds in each group(P>0.05),and the test group needed 20.32±0.31 d to heal,which was significantly lower than the control group’s 23.56±1.05 d healing(P<0.05),which proved that the wound model could heal.Good intra-group stability and inter-group differences;test results of the treatment effect of Ganoderma lucidum spore powder: the average wound healing rate of the experimental group was higher than that of the control group at 3d,7d,14 d,and 21 d,and there was a significant difference at 3d(P<0.05),the test group needed 22.14±1.65 d to heal,which was significantly lower than the control groups 25.06±1.35d(P<0.05).The overall state of the test group was better than that of the control group at 3d and 7d.Section observation showed that the rate of inflammatory cell regression and collagen fiber deposition in the experimental group was better than those in the control group.The results of detecting MDA and SOD showed that the SOD level of the experimental group was significantly higher than that of the control group at 21d(P<0.05).The results of detection of PGE2,TNF-α,and IL-1α showed that the level of IL-1α in the experimental group was significantly higher than that in the control group at 7d(P<0.05),and the level of PGE2 in the experimental group was significantly higher than that in the control group at 14d(P<0.05).The detection of Ig A,Ig M,and Ig G showed that the Ig G level of the experimental group was significantly higher than that of the control group at 7d(P<0.01),and the Ig M level of the experimental group was significantly higher than that of the control group at 14d(P<0.01).The relative expression of PDGF,b FGF,and VEGF detected by real-time PCR showed that the relative expressions of PDGF,b FGF,and VEGF in the experimental group were significantly higher than those in the control group at 7d(P<0.01),and the relative expression of VEGF in the experimental group was significantly higher at 14 d.The expression level was significantly higher than that in the control group(P<0.01).The test results showed that Ganoderma lucidum spore powder can increase the expression of growth factor genes by improving tissue antioxidant activity,inflammatory factors,and immunoglobulin levels.A series of comprehensive effects have a therapeutic effect on dog skin wounds and shorten the wound healing time. |
