| The unique flower heads of Asteraceae plants have important ornamental and research values,and the molecular mechanism of flower development is complex and the related reports are few.Senecio vulgaris belongs to Senecio in Compositae,which has the characteristics of Compositae.At present,the molecular mechanism of flower development of Senecio has yet to be studied.In this study,three MADS-box genes,Sv DEFICINES26(Sv DEF26),Sv DEF162 and Sv GLOBOSA(Sv GLO),were preliminarily obtained by screening the transcriptome data of S.europaea.Bioinformatics analysis was carried out and Sv GLO was selected as the research object.The functions of Sv GLO were studied by real-time quantitative PCR(q RT-PCR)analysis,heterologous transformation of Sv GLO,DNA identification and tissue section analysis.1.The results of bioinformatics analysis show that Open Reading Frame of Sv GLO is 591 bp in length,encoding 196 amino acids,with MADS-box and K-box domains,and PI motif at the c-terminal.2.The results of tissue specificity analysis showed that Sv GLO was strongly expressed only in inflorescence tissues of WT S.vulgaris,but not in vegetative organs.3.During the flower development of S.nigrum overexpressing Sv GLO,it was observed that sepals acquired some characteristics of petals and carpels were transformed into stamen-like organs;The heterologous overexpression of Sv GLO resulted in the increased expression of endogenous B-class genes Sn GLO、Sn PI28、Sn DEF and Sn DEF64 in sepals and carpels of S.nigrum in transgenic plants.Histological staining analysis showed that the morphology of ovary wall cells of transgenic plants was significantly different from that of wild S.nigrum,and the shape and size of anther wall cells of wild S.nigrum were similar.4.During the fruit development period of S.nigrum which overexpresses Sv GLO,the pericarp is brown and cracked,the number of seeds contained in a single transgenic fruit is significantly reduced,and the germination rate of seeds is significantly reduced.The results of this study showed that Sv GLO of S.vulgaris belonged to PI/GLO subfamily genes,and heterologous overexpression of Sv GLO in S.nigrum affected the development of sepals and carpels in transgenic plants.Therefore,it is speculated that Sv GLO may be involved in regulating petal and stamen development in S.vulgaris.The study lay a theoretical foundation for further study on the molecular regulation mechanism of flower development in S.vulgaris. |
