| In this thesis,two lines of wild Lycium ruthenicum Murr.hybrid offspring were used as test materials to establish a regeneration system for the hybrid offspring of two wild Lycium ruthenicum Murr.,the cold resistance of two wild Lycium ruthenicum Murr.crosses was also analyzed.For wild Lycium ruthenicum Murr.hybrid offspring breeding,introduction,development and utilization,provides the experimental and theoretical basis,choose two kinds of wild Lycium ruthenicum Murr.hybrid strains,comparing the branches by measuring conductivity,MAD,POD,SOD,Pro,soluble sugar,soluble protein and other physiological indicators,the comprehensive analysis,in order to compare more suitable for the growth of wild Lycium ruthenicum Murr.hybrid new lines.The regeneration system of hybrid offspring is established,so that it can expand rapidly,and provide technical reference value for the realization of factory seedling raising.The study results are as follows:1.With the original branches of Lycium Ruthenicum Murr.hybrid line 1,the optimal disinfection time for explants is 12 min,the optimal medium for explants induced callus:1/2MS+1.0mg/L 6-BA+0.2mg/L NAA;optimal medium for indefinite bud value: WPM+1.0mg/L 6-BA+0.1mg/L NAA;and optimal medium for root culture: WPM+0.5mg/L IBA.The optimal medium was used as MS hormone-free medium and WPS+0.5mg/L IBA medium.2.Loam + vermiculite + nutrient soil(3:1:1)is the most suitable transplanting substrate for two wild Lycium ruthenicum Murr.hybrid tube seedlings.3.With the decrease of treatment temperature,the relative conductivity of new branches of wild Lycium ruthenicum Murr.crosses increases;MDA content and SS content increase first and then decrease;SOD content and POD content are "down-up-down";Pro content and SS content are "up-down-up-down".Among the seven indicators,the hybrid offspring line 1 was more stable in each physiological index relative to low temperature treatment in line 2.According to the comprehensive evaluation of cold resistance according to the principal component analysis method and membership function method,the hybrid offspring line 1 was greater in cold resistance under cold stress than line 2. |
