| Thymus mongolicus Ronn.is an aromatic ground cover plant widely used in feed,food,medicine,chemical industry and ecological restoration.Thyme is a perennial dwarf semi-shrub,which is distributed in different climatic zones of Ningxia.Wild thyme seeds are difficult to reproduce,and cutting and transplanting are commonly used breeding methods.However,there are few studies on the domestication,cultivation and utilization of wild thyme in Ningxia.In this paper,wild thyme from different areas in Ningxia was taken as the research object,the composition and difference of volatile organic compounds were analyzed,the technical conditions of cutting and transplanting propagation were screened,and the technical methods of transplanting domestication and cutting propagation of wild thyme from different areas were put forward,which provided experimental reference for the development and utilization of wild thyme in Ningxia.The main results are as follows:(1)A total of 339 volatile organic compounds were detected in fresh and dry samples of different parts of thyme from three different areas.Hydrocarbons were the majority(125 species),mainly terpenes p-cymene and y-terpinene,reaching 14.24%-42.24%and 3.3%-37.35%,respectively.The relative content of p-cymene in Nanhuashan thyme was higher than that in Yunwushan and Mahuangshan,and in tender stems was higher than that in lignified stems and leaves,while the relative content in fresh samples and dry samples was equivalent.For the number of volatile components,the number of volatile organic compounds of thyme in Nanhua Mountain was greater than that in Mahuang Mountain and Yunwu Mountain,fresh samples were more than dry samples,and leaves were more than tender stems and lignified stems.(2)Different cutting matrix treatments had significant effects on the rooting and survival of thyme cuttings from different areas.Among them,the survival rate of thyme in Nanhua Mountain and Yunwu Mountain with peat:vermiculite:perlite=1:1:1 was the highest,which was 49.29%and 45.83%,respectively.Different growth regulator types,concentrations and soaking time also have an effect on the rooting and survival of cutting seedlings.Through the orthogonal test design,the semi-lignified thyme was selected as the cutting and soaked in 200 mg·L-1 IBA for 120 min.The survival effect of cutting seedlings was the best.The results of re-cutting of cutting survival seedlings showed that the survival rate of re-cutting with cutting survival seedlings as cutting mother plants was significantly higher than that of direct cutting of wild seedlings.(3)The transplanting test of thyme in the sample area showed that the survival rate of thyme in Yunwu Mountain was 56.56%on the 18 th day of transplanting,which was 1.37 times that of Mahuang Mountain.Shading had a significant effect on the growth and physiology of transplanted wild thyme.The survival rates of Yunwushan and Mahuangshan thyme under 30%shading were 64.05%and 41.60%,respectively,which were significantly higher than those under 55%shading(P<0.05),and were 1.77 times and 1.49 times higher than those under 55%shading,respectively.The branch number,root-shoot ratio and specific leaf area of thyme in Yunwu Mountain and Mahuang Mountain were the highest under 30%shading treatment.Shading 30%is beneficial to the growth of transplanted thyme.In addition,the field transplanting survival rate of thyme cutting seedlings could reach 100%.The plant height and branch number of thyme in Nanhua Mountain were significantly higher than those in Mahuang Mountain and Yunwu Mountain.In summary,wild thyme from different areas in Ningxia has rich volatile organic components and has great application value.In a short period of time,domesticated cultivated plants of wild thyme can be obtained,which can be operated in two steps.Different areas of root thyme were collected,and some of them were transplanted in greenhouse.After survival,they were transplanted to the outdoor field and shaded by 30%.The other part obtains the cutting seedlings through the optimized cutting propagation technology and transplants them to the outdoor field.Both of them can greatly improve the survival rate of field transplanting and obtain the domesticated thyme seedlings.This technical system can be applied and promoted. |
