Research On Screening Of Wheat Germplasm Material For Powdery Mildew Resistance And Molecular Identification Of Resistance Genes

In recent years,the occurrence area and damage of wheat powdery mildew in China have been expanding and aggravation.The resistance genes of the main varieties are single,and they are easy to lose resistance and then lose their use value after years of use.Therefore,it is necessary to cultivate new varieties resistant to powdery mildew constantly.In order to provide a new source of resistance for breeding wheat varieties with persistent powdery mildew resistance,this study combined artificial inoculation and natural disease in the field to observe and screen agronomic traits,identification of resistance at seedling stage and identification of resistance at plant adult stage of 96 materials initially screened in Hangzhou,Zhejiang Province and Kunming,Yunnan Province.And the resistance genes contained in the materials were identified by molecular markers closely linked or co-isolated with known resistance genes.The results show that :(1)The resistance at seedling stage was identified by inoculation with mixed strains in Zhejiang.There were 78 materials showing moderate resistance and above,and 4,24,25 and 25 materials showing immunity,near immunity,high resistance and moderate resistance,respectively.There were 76 materials with moderate resistance and above in Yunnan through natural disease,and 30,37 and 9 materials showed immunity,high resistance and moderate resistance,respectively.There were 67 materials showing resistance at both seedling and adult stages.(2)Nineteen powdery mildew resistance genes were identified by molecular markers.Thirteen resistance genes were detected in 67 resistant germplasm materials,including Pm2,Pm4 a,Pm4b,Pm8,Pm12,Pm18,Pm21,Pm24,Pm26,Pm34,Pm35,Pm57 and Pm Tm4.The number of detected genes was 12,4,14,1,1,1,17,7,2,6,6,3 and2,respectively.Two resistance genes were detected in 11 materials such as“ZW1840”,three resistance genes were detected in 7 materials such as “D23”,and five resistance genes(Pm2,Pm4 b,Pm21,Pm26,Pm35)were detected in “17 Yangfu12 ”.(3)Six varieties(lines)of “Ningmai 1529”,“Ning 1526”,“Guangpengmai 1526”,“ Jinyunmai 3”,“Ningmai 1519” and “ Yangmai 24” were further screened by agronomic traits and yield performance of 67 powdery mildew resistant materials screened from 96 materials in two places,which could be used for further utilization of high yield and powdery mildew resistant wheat breeding in two places.In summary,this study further improved the efficiency of wheat powdery mildew resistance identification,providing a basis for making full use of new resistance sources and selecting excellent green resistant varieties with wide adaptability.
