Most of the fruit trees in the subfamily Liliaceae are self-incompatible,which prevents inbreeding through specific recognition of styles and pollen.Therefore,most of the fruit trees in the subfamily Liliidae need to be cross-pollinated to promote fruit set,and peach is one of the rare fruit trees in the subfamily Liliidae that is self-compatibility.Peach can bear fruit by both self-pollination and cross-pollinated.However,some studies have found that‘Liuyefeitao’exhibits self-compatibility cross-incompatibility during pollination.In this study,we conducted an experimental analysis of the above phenomenon in‘Liuyefeitao’,and utilized the transcriptome sequencing of cross-pollinated,self-pollinated,and unpollinated styles,and screened for the similarity with The transcriptome sequencing of cross-pollinated,self-crossed and unpollinated styles was used to screen for genes related to heterogametic pollination disaffinity in‘Liuyefeitao’,and the differentially expressed genes were bioinformatically analyzed and verified,and the main results of the study are as follows:1、The statistics of self-pollination and cross-pollination fruiting rate of’Liuyefeitao’and the observation test of aniline blue staining of pollen tubes showed that’Liuyefeitao’was self-compatibility but cross-incompatibility.The S genotype of’Liuyefeitao’was identified as S2S2,and sequence analysis revealed that the sequences of LYFT-S2-RNase and LYFT-SLFLs of’Liuyefeitao’were not mutated,while the partial deletion of the pollen sequence of LYFT-SFB2 was the cause of its self-compatibility.q RT-PCR analysis revealed that the expression of LYFT-SLFL3 and LYFT-S2-RNase in the cross-pollinated styles was significantly lower than that in the self-pollinated styles,suggesting that the low expression of LYFT-SLFL3 might be the reason for cross-incompatibility of’Liuyefeitao’,and also suggesting that the low expression of LYFT-SLFL3 might be the reason for the lack of affinity in the pollination hybridization of Liuyefeitao’,and also suggests that non-S factors are involved in the cross-incompatibility of’Liuyefeitao’.2、Transcriptome sequencing was performed on the styles of’WLiuyefeitao’with different pollination treatments for 48 h.The experimental results were analyzed to obtain a total of 5206 differential genes and subjected to KEGG enrichment analysis,among which the phytopathogenic interactions,plant MAPK signaling pathway and plant hormone signaling pathway were significantly enriched,and the expression of calcium-related protein kinases,calreticulin-like(CML),WRKY transcription factors,and growth hormone response factors were significantly expressed differently in the three pathways.3、Calcimodulin-like(CML)was significantly differentially expressed in self-pollination and cross-pollination stylar transcriptome sequencing,and a total of 48CMLs genes,named PpCMLs,were identified in the peach genome data using bioinformatics methods,among which PpCML15 and PpCML24 were significantly higher in expression after heteroflowering than in control and autoflowering,which were hypothesized to be involved in the process of cross-pollination of’Liuyefeitao’as a non-S factor.
