Objective Observing the intervention effect of Banxia Xiexin Decoction on rats with chronic atrophic gastritis(CAG),explore its mechanism through the Notch2/Hes1 pathway,which can provide a molecular basis for the clinical application of Banxia Xiexin Decoction.Methods 68 rats were randomly divided into 2 groups after 1 weeks of adaptability feeding,14 rats in normal group and model group with 54 rats,each half of the male and female.Model group using integrated modeling method,such as deoxycholate sodium,alcohol and active immunization for seventeenth weeks,to establish a model of the chronic atrophic gastritis(CAG);The normal group was treated with equal volume of physiological saline instead of injection and intragastric,drinking water free.After seventeenth weeks modeling,using rats of normal group and model group each 4 rats(each half of the male and female)for pathological examination to ensure that the model is successful.Then the remaining 10 rats of normal group still as the normal group,the model group were randomly divided 50 SD rats into 5 groups:model group,low,medium and high dose groups of Chinese medicine,folic acid group;each group have 10 rats(each half of the male and female).After modeling,begin with eighteenth weeks,normal and model groups were intragastriced the same amount of physiological saline;the chinese medicine groups use of different doses of 6g/kg·d,12g/kg·d,24g/kg·d Chinese medicine compound Banxia Xiexin Decoction,respectively;and the folic acid group was given 5mg/kg·d folic acid,all the groups are for 8 weeks intervention.Pathological changes of gastric tissue were observed by hematoxylin-eosin(HE)staining.Immunohistochemistry was used to detect the expression of proliferating cell nuclear antigen(PCNA)in gastric mucosa of rats in each group,Radioimmunoassay was used to detect serum gastrin in rats,and the expression of Notch2,Jagged1 and Hesl mRNA were detected by Realtime-PCR.Results Using deoxycholate sodium,alcohol with active immunization integrated modeling method for seventeenth weeks can be induced in rats with chronic atrophic gastritis,like gastric mucosa inflammatory cell,the decrease in the number of glands,thinning of the gastric mucosa and so on.compare with the normal rats,CAG rats weight loss evidently(P<0.01),gastric mucosal inflammation score significantly increased(P<0.01),the number of gastric glands and the thickness of gastric mucosal were significantly decreased(P<0.01),the serum gastrin level was significantly decreased(P<0.05),the expression of proliferating cell nuclear antigen of the gastric mucosal was significantly increased(P<0.01),the expression of Notch2,Jagged1 and Hesl mRNA in gastric tissue was evidently up-regulated(P<0.05,P<0.01).After different doses of Banxia Xiexin Decoction and folic acid intervention,comparing the giving different doses of folic acid and Chinese medicine groups with model group,the weight of rats increased evidently(P<0.05,P<0.01),gastric tissue inflammation score decreased significantly(P<0.05,P<0.01),the number of gastric glands and the thickness of gastric tissue were significantly increased(P<0.05,P<0.01),proliferating cell nucleus antigen expression of gastric tissue was evidently reduced(P<0.05,P<0.01),the level of serum gastrin was evidently increased(P<0.05),gastric rats tissue Notch2,Jagged1 and Hesl mRNA expression were evidently lower(P<0.05,P<0.01).Conclusion The occurrence and development of rats’ CAG may be related to the expression of Notch2,Jagged1 and Hesl pathways.Banxia-Xiexin Decoction can prevent CAG progression by interfering with those pathways.
