Study On The Extraction Of Active Ingredients From Shandougen Recipe And Its Anti-Candida Albicans Mechanism

Candida albicans is a gram-positive pathogenic bacteria.It is the most pathogenic in the Candida genus.It exists in the human body under normal conditions and is harmless in symbiosis with the human body.But when the body’s immunity is weakened,or when some immunosuppressive agents such as antibiotics or hormones are used to treat diseases for a long time,Candida albicans can cause infections of the skin,mucous membranes and even the systemic system,and endanger human health.In recent years,due to the rapid development of my country’s medical industry,the use of some immunosuppressants and some hormones has caused the patient’s own immune function to decline day by day.Among these patients with decreased immune function,the risk of Candida albicans infection ranks first.Bit.In this experiment,the traditional Chinese medicine compound Shandougen Recipe was chosen to explore its anti-Candida albicans activity.First of all,this experiment took the minimum inhibitory concentration of the extract against Candida albicans as the final screening index.First,the comparison between water as the solvent and ethanol as the solvent showed that the minimum inhibitory concentration of the extract obtained with ethanol as the solvent Compared with water extraction,the MIC90 of the alcohol extract of Shandougen Recipe is 25 mg/mL.In the next step,the orthogonal design experiment method is used to optimize the extraction process.Based on the single factor experiment,the process optimization of the extraction conditions of Shandougen Recipe is studied.The experimental results show that the best process is the extraction temperature of 85℃ and the number of extractions.Once,the volume fraction of ethanol is 50%,the ratio of material to liquid is 1:25,and the extraction time is 1h.Under these conditions,the minimum inhibitory concentration MIC90 of the extract of Shandougenfang was 6.25 mg/mL.The main chemical components of the ethanol extract of Shandougenfang were determined by UPLC technology.The selected binary gradient mobile phase was acetonitrile and 0.1%phosphoric acid water,and the elution conditions were 0 min,10%acetonitrile;2 min,15%acetonitrile;4 min,21.1%Acetonitrile;12 min;45%acetonitrile,gradient elution is carried out under this condition.The best detection wavelength is 210 nm and the flow rate is 0.3 mL/min.At the same time,methodological investigation was carried out,and the results were all within the normal range.The measured content of matrine was 184.56 ± 2.79 μg/mg,the content of isoferulic acid was 26.67±1.32 μg/mg,the content of cimicifuga was 7.13 ±0.25 μg/mg,the content of shegan glycoside was 10.12 ± 0.61 μg/mg,and the content of irisin was 3.96±0.16 μg/mg.Next,the mice were injected with cyclophosphamide into the abdominal cavity,which inhibited the immune function of the mice,and then injected the Candida albicans spore suspension into the tail vein of the mice,and successfully infected the mice with Candida albicans.The therapeutic effects of the ethanol extract of Shandougen Prescription and its medicinal components were further explored through its administration.The results showed that the Shandougen prescription group and the effective component group can increase the body weight of the mice.The immune organ index of the Shandougen prescription group was significantly higher than that of the model group,and it was found that the ethanol extract of Shandougen prescription It can reduce the growth of Candida albicans in the liver and kidney of mice.In the detection of mouse spleen cell proliferation,it was found that the ethanol extract of Shandougen Recipe and its effective components can promote the proliferation of splenic lymphocytes.Through the scanning electron microscope observation experiment of Candida albicans,it can be seen that the hyphae of Candida albicans in the alcohol extract of Shandougen prescription group and its effective component group were significantly less.After adding glycine,the bacteria became full again.The hyphae also increased.According to this,it can be shown that the alcohol extract of Shandougen prescription and its active ingredients can inhibit the growth of hyphae and have a damaging effect on cell membranes,thereby achieving the effect of treating Candida albicans.
