Objective: To observe the effects of Mongolian medicine moxibustion on body weight,learning and memory,spleen and thymus index,SOD,CAT,MDA index and related transcriptome expression of aging model rats after moxibustion at Dinghui point and kidney point,further clarify the relevant mechanism of Mongolian medicine moxibustion on on aging model rats,and provide objective basis for the anti-aging clinic of Mongolian medicine moxibustion.Methods: Fifty purebred SD rats were randomly divided into five groups: normal group,model group and moxibustion group(Dinghui point,kidney point,Dinghui +kidney point moxibustion group).The rats in the model group and moxibustion group were injected subcutaneously with 5% D-galactose once a day for 42 days.In the normal group,the same amount of0.9% normal saline was injected subcutaneously into the neck once a day for 42 days.During the modeling period,the body weight of each rat was measured and recorded every week,and then the dosage of each rat was recalculated according to the change of body weight.The normal group and the model group were fed normally.The moxibustion group began moxibustion every other week and once every other day,a total of 21 times.Behavioral test:(1)measure the weight once a week and record the weight change.(2)Maze test was used to detect the changes of learning and memory function of rats in each group.(3)Swimming experiment was conducted to detect the physical changes of rats.After the experiment,all rats were anesthetized by intraperitoneal injection of 70% chloral hydrate,and the hippocampus,kidney,spleen,thymus and serum were removed.The indexes of aging related enzymes were measured: MDA,cat and SOD;The indexes of spleen and thymus were calculated;Hippocampus and kidney were frozen in-80 ℃ freezer and transcriptome sequencing was done.Results:The effect of moxibustion on the body weight of aging model rats: there was significant difference between Dinghui point group,Shenxue point group and Dinghui +Shenxue point group and model control group(P < 0.01).Effect of moxibustion on behavioral indexes of aging model rats:(1)The effect of moxibustion on learning ability:there was significant difference between Dinghui group,Shenxue group,Dinghui +Shenxue group and model group(P < 0.01);(2)Effect of moxibustion on memory ability of aging model rats: there was significant difference between Dinghui group,Shenxue group and Dinghui + Shenxue group and model group(P < 0.01)(3)effect of moxibustion on serum indexes of aging model rats:(1)Effect of moxibustion on CAT content: there was significant difference between Dinghui point group and Shenxue point group and model group(P < 0.01).(2)Effect of moxibustion on MDA content: there was significant difference between Dinghui point group,Shenxue point group and model group(P < 0.01).(3)The effect of moxibustion on SOD activity: there was significant difference between Dinghui point group,Shenxue point group and model group(P < 0.01).Through cluster analysis of 1 og2 fol dchange of 199 genes,it was found that C4 B was ubiquitous in three tissues cluster1,which was also demonstrated in other aging related transcriptome sequencing data.In order to obtain the genes with significant tissue-specific changes,we further screened cluster1 according to the conditions of 1og2 foldchange > log(1.5)or1og2 foldchange < log2(0.75).It was found that there were 6 genes in hippocampus and31 genes in kidney.Conclusion: 1.The experimental model is successful,and the 5% D-galactose aging model is suitable for the experimental study on the anti-aging compatibility law of Mongolian medicine moxibustion therapy.(1)The effect of moxibustion on learning ability:there was significant difference between Dinghui group,Shenxue group,Dinghui +Shenxue group and model group(P < 0.01);(2)Effect of moxibustion on memory ability of aging model rats: there was significant difference between Dinghui group,Shenxue group and Dinghui + Shenxue group and model group(P < 0.01)(3)effect of moxibustion on serum indexes of aging model rats:2.It is proved that Mongolian moxibustion therapy uses transcriptome and its signal pathway for anti-aging purpose.3.Mongolian medicine moxibustion therapy can achieve the purpose of anti-aging by affecting the related enzymes(SOD,MDA,CAT),spleen and thymus index,body weight,physical strength,learning and memory ability of aging model rats.
