| Objective:To study the levels of IL-1β/IL-23 in local microenvironment of normal cervical,cervical squamous intraepithelial lesion and cervical cancer tissue,and to analyze their otherness.To analyze the correlation between the expression level of IL-1β/IL-23 in local microenvironment and vaginal microecology and HPV16/18;To study the clinical significance between the expression of IL-1β/IL-23 in cervical cancer tissues,further understand the immune status in cervical lesions.Methods:A total of 96 patients with cervical lesions who underwent surgery in Shanxi Provincial People’s Hospital from September 2019 to September 2020 were selected as the study group,including 28 case of cervical cancer,37 cases of HSIL and 31 cases of LSIL,and 20 cases of uterine excised due to benign diseases(uterine fibroids,etc.)in our hospital at the same time were selected as pair.The exfoliated cells of the vagina and cervix of the patient were collected for examination according to the standard.The vaginal PH,BV and HPV infection results of the subjects were collected.The content of IL-1β/IL-23 was tested by using Immunohistochemical(IHC).Analying the correlation between IL-1β/IL-23 and cervical lesions,and with HPV16/18 infection,vaginal pH,bacterial vaginosis(BV)and cervical lesions.Results:1.In cervical tissue,the rise of likelihood of BV was related to the disease progression compared to the control tissues,P< 0.05,which was statistically significant;Patients with vaginal pH≥4.5 had a higher probability of HPV16/18,P < 0.05;Patients with BV were more likely to be infected with HPV16/18,which had statistically significant(P < 0.05).2.The level of IL-1β/IL-23 expression were significantly different between LSIL,HSIL,CSCC group and control group(P<0.01,ɑ=0.05);There was significant positive correlation between the degree of cervical lesion and the expression of IL-1β/IL-23(rs > 0,P<0.01).The content of IL-23 was different based on different expression levels of IL-1βin cervical tissues(P<0.05);and the correlation was significantly positive of the levels of IL-1β and IL-23(rs > 0,P<0.05).3.Compared with HPV16/18 negative,the tissues had a higher expression levels of IL-1β and IL-23 with HPV16/18,and the expression of IL-1β was mainly positive,which had statistical significance(P<0.05).Compared with BV negative,the expression levels of IL-23 and IL-1β in cervical tissue of vaginal infection BV were higher(P<0.05),there was a statistical difference.4.In cervical cancer tissue,the expression levels of IL-1β and IL-23 were significantly different in different degrees of organizational differentiation and surgical pathological stages(P<0.05),and with the degree of tissue malignancy and surgical pathological stage,the correlation was positive(rs>0,P<0.05).Conclusion:1.In cervical lesions,IL-1β and IL-23 may participate in the occurrence and progress of cervical precancerous lesions.2.IL-1β and IL-23 are related to the pathogenesis of BV,and IL-1β may be an immune cytokine regulating the outcome of HPV16/18 infection.3.In cervical squamous cell caicinoma,IL-1β and IL-23 participate in the proliferation and migration of cancer cells by playing the immunosuppressive role in locally,providing a new direction for the immunotherapy of cervical cancer. |
