Objective:The purpose of this study is to study the inhibitory effect and mechanism of Ganoderma Lucidum Triterpenes on the growth and metastasis of hepatocellular carcinoma from in vivo and in vitro levels,and to investigate whether Ganoderma Lucidum Triterpenes have obvious toxic and side effects.Materials and methods:1.To study the inhibitory effect of Ganoderma lucidum triterpenes on the growth of human hepatoma cell SMMC-7721 in terms of proliferation,apoptosis,migration and invasion phenotypes,and to study the cell cycle,apoptosis and cell proliferation.2.Nude mice SMMC-7721 tumor models were established and divided into three groups.The control group was continuously gavaged with normal saline for 14 days,and the experimental group A(low concentration group)and the experimental group B(high concentration group)were continuously gavaged with Ganoderma lucidum triterpenoid 14 days.Multiple scans of resonance were performed and tumor tissue volume was calculated.3.Detect the liver and kidney function of mice,and perform HE staining on the main parenchymal organ tissues(heart,liver,spleen,lung,and kidney),and perform HE staining and immunohistochemistry(E-cad,Ki-67 and Tunel).Results:1.Ganoderma Lucidum Triterpenoids can inhibit the growth of SMMC-7721 tumor cells at the level of proliferation and apoptosis phenotype in vitro,and inhibit SMMC-7721 tumor cells at the level of migration and invasion phenotype.2.In the in vivo experimental animal model,after statistical analysis,the comparison between the second scan and the third scan of the control group and the experimental group A was statistically significant(p<0.05);the second scan of the control group and the experimental group B The comparison between the scan and the third scan was statistically significant(p < 0.05);and the third comparison between the experimental group A and the experimental group B was statistically significant(p < 0.05).3.The changes of liver and kidney function indexes in nude mouse were negligible,indicating that the treatment had no obvious acute injury to liver and kidney function.HE staining also showed that the main organs(heart,liver,spleen,lung,kidney)in each group had no obvious acute injury or adverse reactions.These results indicated that Ganoderma Lucidum Triterpenoids had no obvious damage to the parenchymal organs of nude mouse.Conclusion:Ganoderma Lucidum Triterpenoids can exert anti-tumor effects by inhibiting tumor cell proliferation,promoting their apoptosis,and inhibiting their invasion and metastasis;they have no obvious toxic and side effects on normal tissues and organs. |
