Objective: Further study on the chemical composition of SAW was carried out to clarify the material basis of its regulation of bone and joint homeostasis Method:(1)The constituents of SAW extract were systematically investigated by using silica gel,ODS,gel Sephadex LH-20,and other chromatographic materials,combined with column chromatography,preparative thin layer chromatography,and semi-preparative liquid chromatography.Modern spectroscopic means such as NMR,MS,and IR were used to identify the chemical structures of the obtained compounds(2)To study in vitro activities of compounds 31 and33 isolated from the ethyl acetate extract of SAW,EA.hy926 cells were used as a model,and six different concentrations of compounds 31 and 33 were given for intervention,and the cytotoxicity of the tested substances was tested at 24 h and 48 h,respectively.Furthermore,cell viability was used to evaluate the effect of monomer compounds on EA.hy926 cells.Result:(1)52 compounds were isolated from the extract of SAW,including a new compound 1*,fifteen pentacyclic triterpenes(2-16),eight steroids(17-24),six flavonoids(25-30),four iridoids(31-34),one cyanide(35),three straight-strand diterpenes(39-41),three alkaloids(43-45),two fatty acids(51-52),nine other type compounds(36-38,42,46-50).(2)Compound 31 had almost no effect on the survival rate of EA.hy926 with the increase of concentration,while compound 33 had a greater effect on the survival rate of EA.hy926,according to the dose tolerance.In addition,compared with the normal group and the model group,compound 31 had a more obvious inhibitory effect on cell tube formation,while compound 33 had no obvious inhibitory effect.Conclusion: 52 compounds were isolated from the extract of SAW,among them,compound 1* was isolated as a new compound,2,5-7,9-11,14,17-24,were isolated from this genus for the first time,and 25,26-50 were all isolated from this species for the first time.Among them,compound 31 isolated from this plant for the first time can inhibit the tube formation of EA.hy926 cells in vitro,and has the potential to inhibit angiogenic activity in vitro,but the mechanism is not clear and needs further study.The results of this paper provide a reference for clarifying the material structure of SAW in regulating bone and joint homeostasis.
