| Torreya grandis Fort.ex Lindl cv.Merrillii Hu is a good variety of Torreya grandis,Torreya genus in Taxaceae,which was artificially bred.Chinese Pharmacopoeia believes that it has the effect of killing insects and eliminating accumulation,moistening lung and relieving cough,moistening dryness and relieving constipation.With the continuous in-depth research on T.grandis,it has been found that T.grandis has antibacterial,anti-tumor,antiinflammatory,hypolipidemic and other biological activities.In this paper,the aril of T.grandis was used as the research object.The diterpenoids chemical components of T.grandis were extracted,separated,identified,and their biological activities were evaluated.The main results are shown as follows:(1)30 compounds were isolated by various chromatographic method(silica gel,bonded silica gel,Sephadex LH-20,HPLC,etc.).The structures of all the compounds were identified based on various spectral analysis methods(1D/2D NMR,CD,MS,X-Ray,etc.).The 30 compounds were 26 abietane-type diterpenoids,3 labdane-type diterpenoids and 1 cadinanetype sesquiterpenoids.There were 9 new compounds and 2 new natural products among 26abietane-type diterpenoids.(2)Some compounds and T.grandis extracts were evaluated for their antibacterial activity,cytotoxic activity and anti-neuroinflammatory activity.The results of antibacterial activity test showed that 19 and 20 had significant inhibitory effects on methicillin-resistant Staphylococcus aureus(with MIC values of 28.6 and 30.0 μg/m L,respectively);The petroleum ether extract,ethyl acetate extract and J5 components of T.grandis extract had certain inhibitory effects on MRSA(with MIC values of 25,100 and 25 μg/m L,respectively).The results of cytotoxic activity test showed that all the compounds had no cytotoxic activity against hepatoma cells HepG2 at 20 μM concentration.The results of antineuroinflammatory activity test showed that 20,27 and 28 had significant antineuroinflammatory activity,the inhibition rate of NO release by LPS-induced BV-2microglia cells were more than 70% at 20 μM concentration,and the cell survival rate was more than 90%. |
