Torreya grandis cv.merrillii is the only grafted and breeded thoroughbred specie of Torreya grandis Fort.ex Lindl.which is a genus from the family Taxaceae and category Torreya.Torreya grandis cv.merrillii has a cultivation history for more than 1300 years,is a rare and unique species mainly distributed in the hilly areas of subtropical China,particularly in Zhejiang,Jiangsu,Anhui provinces.the succulent seed aril that completely encloses the seeds and represents about 50%of the fresh weight of the seeds,is normally discarded which consequently resulted in great waste of resources and generated some environmental problems.In this paper,torreya grandis aril was used as the researching object.Its chemical components and bioactivities were analyzed.The mine results were as follows:1.Extraction and separation of the chemical constituents of torreya grandis arilThe ethanol site of Torreya grandis aril was extracted with ethyl acetate,petroleum ether,N-butanol respectively.Adopts the chromatography like silica gel etc,to make a study on the chemical components of torreya grandis aril and determines its compound structures according to their physiochemical property and the spectrum technology.And the following nine compounds are abtained:Torreyagrandatc?Compound 1?,Hinokiol?Compound 2?,?-Sitosterol?Compound3?,3,4-Dihydroxybenzoic Acid 3-O-?-D-Glucuronide?Compound 4?,4-epiagathadial?Compound5?,dehydroabietic acid?Compound 6?,Trans-communic acid?Compound 7?,Cis-communic acid?Compound 8?,2-methoxy-1,4-phenylene)dimethanol?Compound 9?.2.The determination of total flavonoids and total phenolic acids content of torreya grandis arilA content determination of the effective components has been made in torreya grandis aril:total flavonoids and total phenolic acids measured by NaNO2-Al?NO?3 method and Folin-Ciocalteu method.The results show that the content of total flavonoids and total phenolic acids were different in different solvent fractions of Torreya grandis ail,in which the most content of total flavonoids and total phenolic acids was n-butanol fraction of Torreya grandis ail.3.Study the biological activity of torreya grandis aril in vitroFor the different extraction parts of torreya grandis aril,the paper adopts the adopts the methods of DPPH,FRAP,and total reducing power,and by determining their IC500 value and TEAC value to make an assessment on their antioxidant activity;adopts the lethal-to-prawn bioactivity experiment and by determining the LC500 value to make an assessment on their cytotoxicity.The results show that the different extraction parts of torreya grandis aril all have certain antioxidant activity and cytotoxic activity.N-butanol fraction of torreya grandis aril shows the strongest antioxidant activity and cytotoxic activity with LC50 is 227.8?g·mL-1.4.Study the biological activity of torreya grandis aril in vivoThis paper also makes an experimental study on the anti-tumor effects on H22 mice models of liver cancer of n-butanol fraction of torreya grandis aril in vivo.60 ICR male mice were used to establish H22 mice models and then randomly divided into six groups,The normal control group,the model control group,the positive group?cyclophosphamide:CTX?,the administration groups(High,4g·kg-1;Medium,2 g·kg-1;Low,1 g·kg-1).The animals were sacrificed 1month after oral administration and tumors were taken out for the tumor weights and anti-tumor rates,while thymus and spleen were taken for thymus index and spleen index.Blood in eyeball was collected for the determination of tumor related indicators in serum.Sections of tumor issue were prepared and morphological changes in tumor tissue cells were observed by using hematoxylin and eosin?HE?staining technique.The results show that n-butanol fraction from aril of torreya grandis can potentiate immunologic function and antioxidant activity of tumor-bearing mice and show out anti-tumor activity without obviously liver damage.
