Torreya grandis cv.merrillii is belong to the Torreya genus of Taxaceae family which has been cultivated with a long history.There are many variations of Torreya grandis Fort.ex Lindl and Torreya grandis cv.merrillii is the best variation type and the unique cultivars of which.It has many development values due to its abundant biologically active ingredients and pharmacological activities.This paper focuses on the research of chemical constituents and biological activities of Torreya grandis leaves,which can provide references for the development and resources utilization of Torreya grandis.The experimental results are as follows:1.Determination of the contents of total flavonoids and total phenolic acid in different extract fractions of Torreya grandis leaves.The content of total flavonoids and total phenolic were measured by NaNO2-Al(NO3)3 method and Folin-Ciocalteu method.The results showed that there are flavonoids and phenolic acids in different extracts of 70% ethanol extract of Torreya grandis leaves and the content of total flavonoids and total phenolic acid of ethyl acetate is the highest.2.Isolation and identification of chemical constituents of ethyl acetate and n-butanol extract fractions of 70% ethanol extract of Torreya grandis leaves.Torreya grandis leaves ethanol site extract respectively with ethyl acetate,petroleum ether,n-butanol.Separated by various column chromatography,chemical methods and technologies pop were used to identify the 11 compounds,respectively: 3,4-two hydroxy benzoic acid(Compound 1),pisifera phenol(Compound 2),catecho(Compound 3)l,Torreya ester(Compound 4),3 4-,two hydroxy benzoic acid methyl ester(Compound 5)4-(3-Methoxy-allyloxy)-benzene-1,2-diol(Compound 6),(7,8-cis-8,8'trans)-2',4 'two 5 two hydroxy-3 methoxy-lariciresino(Compound 7),Acrylic acid 3-phenyl-allyl ester(Compound 8),?-sitosterol(Compound 9),Cis-communic acid(Compound 10),Trans-communic acid(Compound 11).3.Study the biological of Torreya grandis leaves in vitro and in vivo.To determine the antioxidant activity of Torreya grandis leaves 70% ethanol extract.The DPPH assays FRAP and total reduction ability were used for determining both Torreya grandis leaves70%ethanol extract.Ethyl acetate extract antioxidant activity is best.The n-butanol extract had certain antioxidant activity.Antioxidant activity of petroleum ether extract is the least significant.A total of 60 male ICR mice were randomly divided into 5 groups: control group(saline),positive group(Red Bull beverage),70% ethanol extract of Torreya grandis leaves(Low:0.8 g/kg,Medium:1.6 g/kg,High:3.2 g/kg)dose groups,and were oral administration of 14 d continuously.At last after treatment,all the mice were forced to weight load exhaustive swimming for 30 min.Then,the contents of lactic acid and urea nitrogen in serum,liver glycogen and muscle glycogen were determined by ELISA.The results show that compared with the control group,70% ethanol extract of Torreya grandis leaves can significantly improve the liver glycogen and muscle glycogen,reduce theserum lactic acid,urea nitrogen and the content of MDA,prolong the mice swimming time,has obvious anti fatigue effect,especially in the high dose group.It provides a reference for the rational development of resources in-depth application of Torreya grandis leaves.
