| Objective To evaluate the efficacy and biological safety of in situ electrospinning technology based on a new type of medical glue for rapid hemostasis after abdominal aorta trauma in rats.Methods The scanning electron microscope(SEM)was used to find out the parameters of the electrospun instrument with the best morphology of the medical rubber fiber,and then the mechanical properties of the electrospun membrane were evaluated by mechanical explosion test and pig lung air leakage test in vitro under this condition,and the biological safety of the electrospun membrane was evaluated by hemolysis test and cytotoxicity test,Finally,the model of tail amputation hemostasis in mice(divided into four groups: blank group,10 s block group,10 s electrospun group,30 s electrospun group)and the model of abdominal aorta wound and hemorrhage in SD rats(divided into three groups: traditional suture group,smearing group and electrospun group)were established.The former observation index was the total amount of bleeding within 5 minutes,and the latter observation index included: hemostasis time,intraoperative bleeding amount,change value of vascular diameter,and success rate of first hemostasis,Doppler ultrasound was performed one week after operation,and histopathology was taken 28 days after operation.Results The spinning effect of the medical glue was good when the electrospinner was adjusted at a speed of 20-25ul/min,the voltage was 14-18 kv,and the distance was 6-10 cm.The in vitro experiment showed that its biological safety was good.In the mouse tail amputation model,the electrospun group can achieve complete hemostasis within 30s;In the model of abdominal aorta trauma and bleeding in rats,all animals survived after operation.Compared with the suture group,the electrospinning group had less bleeding(P<0.01),smaller absolute value of vessel diameter change(P<0.01),and less intimal hyperplasia(P<0.01);Compared with the smear and suture groups,the electrospun group had shorter vascular closure time(P<0.01)and fewer inflammatory cells(P<0.05)Conclusion The nanofibers prepared by in situ electrospinning technology based on the medical glue can quickly seal the covered artery wound to achieve hemostasis,which is more timesaving than the suture method,less inflammatory than the application method,and lower the rate of late vascular stenosis. |
