| Sophora tonkinensis Gapnep.is the dried rhizome and root of Vietnamese acacia,mostly distributed in Guangdong,Guangxi,Jiangxi and other places.It has the efficacy of clearing heat and detoxification,reducing swelling and throat,and is often used for fire and poisonous accumulation,milk moth throat paralysis,sore throat,swollen gums and sores on the mouth and tongue.The dried rhizome of Menispermum dauricum DC.,which can clear heat,detoxify,dispel wind and relieve pain,can be used for sore throat,diarrhea and dysentery caused by fever and rheumatism.Network pharmacology and molecular docking were used to analyze Sophora tonkinensis Gapnep.and Menispermum dauricum DC.,and the extracted active ingredients were verified by biological experiments.The extracted active ingredients were analyzed by using network pharmacology and molecular docking for the analysis of Sophora tonkinensis Gapnep.and Menispermum dauricum DC.,and the extracted active ingredients were validated by biological experiments.By using liquid chromatography-mass spectrometry,we established a method for the determination of the active ingredient content in Sophora tonkinensis Gapnep.and Menispermum dauricum DC.to provide a reference for the quality evaluation of the two herbs.Some of the active ingredients were analyzed by mass spectrometry for the cleavage pathway.From the analysis of the "component-target-disease" network diagram of ophora tonkinensis Gapnep.,it can be seen that quercetin and kaempferol,the two components related to the most targets,are the main active components of ophora tonkinensis Gapnep..The active ingredients related to PPARG,PTGS2,AR,and HSP90 were the main targets affecting the efficacy of ophora tonkinensis Gapnep..There are more targets related to lung cancer,leukemia,non-selective cancer,breast cancer,liver cancer,and bacterial infection.Analytical studies on GO bioprocess enrichment and KEGG metabolic pathway enrichment showed that santoprene can inhibit cancer and inflammation.Molecular docking showed that PTGS2 was an important target for the effect of ophora tonkinensis Gapnep.,and quercetin and kaempferol were important active ingredients of ophora tonkinensis Gapnep..The best extraction conditions of ophora tonkinensis Gapnep.were achieved by using 75%methanol solution as the extraction solvent with a material to liquid ratio of 1:10 and reflux for 2h.The extract of ophora tonkinensis Gapnep.showed inhibitory effects on A549,Hela,Hep G-2,RAW,MCF-7,Staphylococcus aureus and Escherichia coli.The cleavage pathways of locustine,rhodopsin,quercetin and auriculine were analyzed and α cleavage was predominant.The analysis of the network diagram of "component-target-disease" in the study of Menispermum dauricum DC.showed that batrachydrine and levulinic acid had the most relevant targets and were the main active components of Menispermum dauricum DC..The targets related to SCN5 A,PTGS1 and AR were more frequent and were the main targets affecting the efficacy of Menispermum dauricum DC..There are more targets associated with leukemia,breast cancer,with non-selective cancer,lung cancer,and bacterial infection.The analysis of GO biological process enrichment and KEGG metabolic pathway enrichment showed that the ability of Menispermum dauricum DC.can inhibit cancer and inflammation.The results of the docking of the active ingredients of the root with the relevant targets showed that PTGS2 and TOP2 were important targets for the action of the active ingredients of the root,and the alkaloids of the root were important active ingredients.The best extraction protocol was to use 75% methanol solution as the extraction solvent,with a material-liquid ratio of 1:10 and reflux for 2h.The extract showed inhibitory effects on MCF-7,RAW,A549,Hep G-2 and Hela.The degree of inhibition of Staphylococcus aureus and Escherichia coli by northern bean root extract increased with time and concentration.The cleavage pathway analysis of cyanophylline and epiberberine was dominated by alpha cleavage. |
