| Objective To study the effect of acupuncture on mammary gland hyperplasia in rats by i TRAQ technique,taking the meridian disease of stomach meridian of foot-yangming as the carrier of disease research and taking the stomach meridian of foot-yangming as the different state of body as the breakthrough point.Methods Twenty rats were randomly selected and divided into model group(n=10)and acupuncture group(n=10).Another 10 normal female SD rats were selected as blank group.The rats in the acupuncture group were treated with acupuncture at Zusanli(ST 36),Liangqiu(ST 34)and Xiajuxu(ST37)of the stomach meridian of Foot-yangming according to the method of "selecting acupoints along meridians",30 minutes each time,once a day,10 times in total.The other two groups did not receive experimental intervention.The height and diameter of the nipple were measured by electronic vernier caliper in each group.The pathological changes of the rat mammary gland were observed by HE staining.The pathological changes of the lesion cells in the mammary tissue of the rats before and after acupuncture intervention were observed.Results 1.Before acupuncture intervention,the height and diameter of papilla in acupuncture group were significantly increased(P<0.05);The serum E2 level was increased and the serum P level was decreased(P<0.05).HE staining showed that the number of acinar lumens and ducts in the milk of the model group increased and the acinar epithelium appeared hyperplasia.After acupuncture intervention,the nipple height and diameter in the acupuncture group were significantly reduced(P<0.05).The serum E2 level was decreased and the serum P level was increased(P<0.05).HE staining showed that the number of acini in the breast decreased,the acini atrophied,and the secretion in the glandular lumen decreased.2.Screening and identification of differential proteins in breast tissue of rats in each group:(1)4655,5126 and 5110 different proteins were identified in blank group,model group and acupuncture group,respectively;Compared with model group,218 proteins in blank group were up-regulated and 243 proteins were down-regulated.Compared with the acupuncture group,52 proteins in the model group were up-regulated and 38 proteins were down-regulated.(2)Four different proteins were found in blank group and model group,and in model group and acupuncture group.3.Enrichment analysis of differentially expressed proteins in each group:(1)GO function analysis showed that the differentially expressed proteins in the model group and the blank group were mainly involved in sarcomere organization,myofibril assembly and voluntary skeletal muscle contraction;The differential proteins between the acupuncture group and the model group were mainly involved in Fc Rn immunoglobulin receptor-mediated Ig G immunoglobulin transport in epithelial cells,antigen processing and TAP-dependent presentation of endogenous peptide antigen through MHC class I through the endoplasmic reticulum pathway,and processing and presentation of exogenous peptide antigen through MHC class Ib.It was found during the analysis of cellular localization.The differentially expressed proteins between the model group and the blank group were mainly located in myocardial myofibrils,M-band and sarcomere.The differential proteins between the acupuncture group and the model group were mainly located in MHC class I protein complex,MHC class I peptide-loading complex,and hydrogen: potassium exchange atpase complex.In the process of molecular function analysis,it was found that the differentially expressed proteins between the model group and the blank group were mainly involved in muscle structure,G protein-activated inward rectifying potassium channel activity and sphingosine N-acyltransferase activity.The differential proteins between the acupuncture group and the model group were mainly involved in TAP2 binding,β-2-microglobulin binding and TAP binding.(2)KEGG analysis showed that in the model group compared with the blank group,differential proteins were mainly concentrated in glycolysis/gluconeogenesis,amino acid biosynthesis and other pathways;in the acupuncture group compared with the model group,differential proteins were mainly concentrated in autoimmune thyroid disease,allograft rejection,graft-vers-host disease and other pathways.(3)PPI results showed that IMP4,PNO1,GNL3 L were the main different proteins in the model group compared with the blank group.CREBBP,ACTG2,SEC61A1 and so on were found in the acupuncture group compared with the model group.Conclusion The effect of acupuncture on hyperplasia of mammary gland is obvious,and it has obvious superiority in apparent,pathological and biochemical indexes.Through proteomic analysis,the treatment of mammary hyperplasia by acupuncture "following the meridian and selecting points" is a holistic regulation of the body through multiple systems and multiple pathways,so as to treat mammary hyperplasia.The possible mechanism is that:(1)the activity and metabolism of phospholipid and sphingolipid are inhibited by up-regulation of PDXDC1 protein,thus inhibiting the hyperplasia of mammary gland.(2)The important genes that inhibit breast hyperplasia factor are activated by up-regulating METTL9;(3)The neuroinflammatory state around the mammary gland was changed by down-regulating LYZ2.(4)By downregulating LARP 1 and binding with other substances in the cell as a cofactor,RNA degradation can cause the division and apoptosis of mammary hyperplasia cells,so as to treat mammary hyperplasia. |
