| Although mineral elements only account for circa 1/10,000 of the total mass of the animal body,they are widely involved in the body’s composition,metabolism,immunity,and growth,and play an important role in the physiological activities of the animal body.Different mineral elements are involved in life activities in different manners,but in most cases,they are intertwined to maintain the operation of this huge and complex biological organism.Animals cannot synthesize mineral elements on their own but by diets.Under grazing conditions or certain specific growth stages of animals,unscientific nutrient supply may lead to an imbalance in the homeostasis of mineral elements,resulting in stunted growth,reduced resistance to stress,impaired immune function and other problems,which will bring economic losses to animal husbandry.To study the effect of sheep mineral element homeostasis imbalance on leukocyte calprotectin and serum immune factors,45 two-month-old female Small Tail Han sheep with similar body condition were randomly divided into a house feeding group(22)and a grazing group(23)in this experiment.The trial was carried out in Zhaozhou County,Daqing City,Heilongjiang Province for5 months.After the trial period,blood sample was collected through the jugular vein and serum and leukocytes were separated.The concentration of seven mineral elements of calcium(Ca),phosphorus(P),potassium(K),magnesium(Mg),iron(Fe),zinc(Zn),and copper(Cu)in sheep serum was detected by kits;The relative expression levels of calprotectin family S100A8 m RNA,S100A9 m RNA and S100A12 m RNA in sheep leukocytes were detected by fluorescence real-time quantitative PCR;The concentration of IL-6,IL-8,TNF-α,IgG,S100A8,S100A9 and S100A12 in the serum of the sheep was detected by an enzyme-linked immunosorbent assay(ELISA).The relationship between mineral elements and between mineral elements and leukocyte calprotectin and serum immune factors was analyzed after mineral element homeostasis imbalance in sheep.The test results showed that the concentrations of mineral elements Ca,P,and Cu in sheep serum did not change much under different feeding conditions(P>0.05).The relative expression level of K concentration decreased(P<0.01),Fe concentration decreased(P<0.001),Zn concentration increased(P<0.05),S100A8 m RNA(P<0.05),S100A9 m RNA(P<0.05),S100A12 m RNA(P<0.05)in leukocytes increased significantly,while the concentrations of SA100A8,S100A9,S100A12,IL-6,and TNF-α in sheep serum under different feeding conditions did not change much(P>0.05).The concentrations of IL-8 and IgG were increased(P<0.05).The results of correlation analysis showed that in sheep serum under grazing conditions,the mineral element Fe was positively correlated with Mg(R=0.452,P<0.05);Fe was negatively correlated with Cu(R=-0.509,P<0.05);Ca was positively correlated(R=0.584,P<0.01);K was positively correlated with S100A8(R=0.428,P<0.05);K was positively correlated with S100A9(R=0.555,P<0.01);Fe was positively correlated with IgG(R=0.504,P<0.05);S100A8 was positively correlated with S100A9(R=0.809,P<0.01);S100A8 was positively correlated with IL-6(R=0.639,P<0.01),IgG,(R=0.597,P<0.01),TNF-α(R=0.507,P<0.05)was positively correlated;S100A9 was positively correlated with IL-6(R=0.701,P<0.001),IL-8(R=0.583,P<0.01),IgG(R=0.580,P <0.01),TNF-α(R=0.780,P<0.001)were positively correlated;S100A12 was negatively correlated with IL-6(R=-0.464,P<0.05).To sum up,the mineral element homeostasis of grazing sheep in this region was unbalanced;the calprotectin S100A8,S100A9,and S100A12 m RNA expressions in leukocytes of sheep with unbalanced mineral element homeostasis increased,but the difference in serum calprotectin concentration was not significant statistically.The serum K concentration was positively correlated with the serum S100A8 and S100A9 protein concentrations;the serum IgG and IL-8concentrations of the sheep with unbalanced mineral element homeostasis increased,and the serum Fe concentration was positively correlated with the serum IgG concentration. |
