| Objective:This study evaluated the safety of the new dendrobium officinale decoction through the acute skin toxicity,long-term toxicity and oral mucosa irritation experimental research of dendrobium officinale decoction on mice,and laid the foundation for the later toxicity test of related preparations of Dendrobium officinale decoction in order to promote the potential value of Dendrobium chinense in the treatment of skin diseases.Methods:1.Skin toxicity test :30 C57 bl / 6J mice were randomly divided into blank control group,intact skin high-dose group,intact skin low-dose group,damaged skin high-dose group,damaged skin low-dose group,a total of 5 groups,6 mice in each group,male and female half,both sides of the back spine of mice were depilated.(1)Acute skin toxicity test: the new dendrobium officinale decoction or normal saline was directly applied to the intact and damaged skin in the depilation area of mice.The erythema and edema on the skin of the administration site were observed for 7consecutive days.Meanwhile,the body weight,diet,behavioral activity,nervous system,respiratory system,gastrointestinal system,hair,mucosa and other poisoning manifestations and death of the mice were observed.(2)Long-term skin toxicity test:The tested drug was applied externally to intact and damaged skin in the hairless area of mice for 4 h,twice a day for consecutive 90 days.Erythema and edema on the skin of the administration site were observed,and body weight was measured every 4weeks.24 h after the last administration,blood samples were taken and each group was sacrificed for observation of blood,blood biochemistry,viscera anatomy and histopathology.2.Mucosal irritation test: mice were randomly divided into intact mucosa group and damaged mucosa group.The left and right sides of their own mucosa were compared.24 hours and 7 days after oral mucosa administration,the area was observed for hyperemia,edema and other irritation phenomena.72 h after the last administration,the animals were sacrificed,and the oral mucosa stimulation response was observed by naked eye and histopathological examination.Results:(1)Acute skin toxicity test: After 24 hours of administration,continuous observation for 7d,no erythema,edema and other stimulation reactions were found in intact or damaged skin of mice caused by Dendrobium chinense decoction or normal saline.No abnormal behavior,activity,diet and other general conditions were observed in each group,and no death was found in each group.T test showed no statistically significant difference in body weight between groups(P> 0.05).(2)Longterm skin toxicity test: 90 days after long-term administration,there were no significant abnormalities in the behavioral activities,general conditions such as body weight,blood biochemistry and other indexes of the mice,as well as the indexes of the main tissues and organs,and no significant toxicity in the main organs.There were no statistical differences in the observed indexes between groups by one-way ANOVA(P> 0.05),there was no significant difference in weight gain between groups by repeated measures ANOVA(P>0.05).(3)Oral mucosa irritation test: After repeated administration of oral mucosa in mice for 7 days,no obvious erythema,edema and other stimulation reactions were found in the mucosa,the irritation score was "0",and no abnormality was found in histopathological examination.Conclusion:(1)There was no acute skin toxicity of Dendrobium chinense.(2)There was no significant long-term skin toxicity of Dendrobium chinense.(3)Dendrobium chinense had no obvious mucosal stimulation reaction and no irritation to oral mucosa. |
