| ObjectiveTo evaluate the clinical effectiveness of scalp acupuncture in acute ischemic stroke by Meta-analysis,and to observe the effects of combined scalp acupuncture with body acupuncture on neurological function scores and STAT3 and p-STAT3 protein expression in brain and small intestine tissues of rats with acute focal cerebral ischemia,and to explore the mechanism of action of acupuncture in the treatment of acute ischemic stroke and preliminary evidence of encephalopathy and intestine.Method(1)Literature Research:Meta-analysis of the input data was performed with Review Manager software,and the analysis results were visualized with Meta View in the form of charts,especially in the form of forest plots,to sort out the intervention status of relevant indicators.(2)Basic research:Eighteen SPF-grade male SD rats were selected and randomly divided into three groups:flase operation group(group A),model group(group B),and cephalic plexus combined with body acupuncture group(group C),with six rats in each group.group A prepared the flase operation model,and groups B and C prepared the right-sided focal cerebral ischemia(MCAO)model using the Zlonga modified wire embolization method.Groups A and B were only grasped without therapeutic intervention,and group C was intervened by cephalic plexus stabbing combined with body acupuncture.The rats in each group were subjected to Zea-longa neurological function score at 4 h and 14 days after the postoperative treatment intervention;the expression of STAT3 and p-STAT3 proteins in brain and small intestine were detected by immunofluorescence and immunohistochemistry techniques at the end of 14 days of treatment intervention in each group,and the effects of scalp acupuncture combined with body acupuncture on neurological function score and on STAT3 and p-STAT3 protein expression in MCAO rats were observed.p-STAT3 protein expression in MCAO rats,and to investigate the mechanism of acupuncture in the treatment of ischemic stroke.Results1.Literature Research:Meta-analysis included 10 studies with 958 subjects and showed that the addition of head injection to baseline treatment reduced neurological function scores,improved levels of daily living ability and increased overall clinical effectiveness in patients with acute ischemic stroke.2.Basic research:(1)Neurological function scores:4h postoperatively,the difference was statistically significant(P<0.05)in group A compared with groups B and C;there was no significant difference in group B compared with group C(P>0.05).After 14 days of intervention,group C decreased significantly compared with group B,and the difference was statistically significant(P<0.05)(2)Expression of STAT3 and p-STAT3 in brain tissues:after 14 days of intervention,the average fluorescence intensity results showed an increasing trend of STAT3 and p-STAT3 protein in group B compared with group A;group C was compared with group B,which showed a decreasing trend;the average optical density showed an increasing trend of STAT3 and p-STAT3 in group B compared with group A;group B was compared with group C,which showed a decreasing trend.(3)Expression of STAT3 and p-STAT3 in small intestine tissues:the mean fluorescence intensity results showed that the expression of STAT3 was significantly increased in group B compared with group A,and the difference was statistically significant(P<0.05),and the expression of p-STAT3 showed an upward trend.Comparing group B with group C,STAT3 protein was significantly decreased in group C,and the difference was statistically significant(P<0.05),and p-STAT3 showed a decreasing trend compared with group B,but it was not statistically significant(P>0.05).The mean optical density results showed that the expression levels of STAT3 and p-STAT3 in group B increased significantly compared with group A,and the difference was statistically significant(P<0.05);when group B was compared group C,the expression levels of STAT3 and p-STAT3 in group C showed a decreasing trend compared with group B,but the difference was not statistically significant(P>0.05).Conclusion1.Literature Research:The results of the Meta-analysis proved that scalp acupuncture added to baseline treatment is an effective method for the treatment of acute ischemic stroke,which can improve the overall clinical efficiency,reduce neurological deficits and improve patients’ ability to perform daily life.2.Basic Research:The expression of STAT3 and p-STAT3 in the small intestine of MCAO rats increased significantly at 14 days after acute ischemic stroke,which may preliminarily confirm the existence of encephalopathy and intestine.Early interventional acupuncture treatment after acute ischemic stroke can effectively improve neurological deficits,and its mechanism may be achieved by inhibiting the expression of STAT3 and p-STAT3 in small intestine tissue. |
