Objective: To study the inhibitory effect of Longdan Xiegan decoction on HSV-2 by network pharmacology and proteomics,and to explore the mechanism of Longdan Xiegan decoction in treating genital herpes.Methods:1.The effective targets of Longdan Xiegan decoction against HSV-2 infection were obtained by using TCMSP,Gene Cards,Dis Ge NET,OMIM and other databases.The mechanism of action was predicted by GO and KEGG enrichment analysis.2.The SH-SY5 Y cell model infected by HSV-2 was established and divided into groups.The cell protein was extracted from the serum containing Longdan Xiegan decoction,label-free quantitative proteomics was used to analyze the protein quantitation and significance difference of the samples,and the most significant enrichment pathway was selected by GO and KEGG annotation and enrichment analysis,the mechanism of action was analyzed and predicted.Results:1.There are 179 effective chemical components in Longdan Xiegan decoction,which correspond to 245 related targets.The main active components are quercetin,kaempferol,stigmasterol,β-sitosterol and isorhamnetin,there are 39 common targets between GDT and HSV-2.The key targets are RELA,MAPK3,IL-6,MAPK1,TP53,TNF,STAT1,IL-10,IL-4,IL-1β,the anti-infection of HSV-2 is mainly through affecting the regulation of cell proliferation and apoptosis,inflammatory reaction,protein binding,TNF receptor binding,MAPK activity,viral receptor activity.2.The proteomic results showed that there were 709 differentially expressed proteins in the course of anti HSV-2 infection,of which 415 proteins were up-regulated and 294 proteins were down-regulated,through enrichment analysis,it was found that these proteins were mainly concentrated in the regulation of m RNA stability,MAPK cascade,ATP binding,cell cycle,metabolic pathway.Conclusion:1.The main active ingredients of Longdan Xiegan Decoction on HSV-2 are quercetin,kaempferol,stigmasterol,β-sitosterol,isorhamnetin,etc.2.Longdan Xiegan Decoction mainly acts on the MAPK signaling pathway through the core targets such as MAPK3,IL-6,IL-10,TNF,etc,inhibits the replication of HSV-2 virus in cells,and regulates the body’s immune function,so as to achieve the role of treating genital herpes.
