Objective:Exercise can slow down the occurrence and development of chronic cardiovascular diseases,and has a certain therapeutic effect on early chronic cardiovascular diseases in suitable populations,but whether it can change or even improve the balance of collagen substances in aortic cells to prevent and treat chronic cardiovascular diseases is not completely clear.In this study,the effect of one-time HIIT on aortic matrix metalloproteinase-2 and related regulators in adult rats of different months of age was observed,and it was explored whether exercise could affect cell processes by triggering inflammatory responses and thereby improve the collagen composition of rat aortic vessels,which provided an important theoretical basis for making up for the gap in stable,advanced and safe non-drug means for the prevention and treatment of chronic cardiovascular diseases.Methods:This study is a one-time HIIT exercise experiment.In the experiment,8-week-old adult SD rats and 8-9 months old SD rats were recorded as youth group(C group)and adult group(Z group),and were divided into two large groups,exercise group and experimental group,respectively.Among them,two quiet groups did not have any intervention,and were recorded as CR group and ZR group.The two exercise groups were subjected to a one-time HIIT exercise experiment,and were divided into four groups immediately after exercise,3h,12h and 24h,which were recorded as OCH group,3CH group,12CH group,24CH group and 0ZH group,3ZH group,12ZH group and 24ZH group.The Western Blot method was used to express MMP-2,P4Hα1,p21,Cyclin D1 and TNF-α proteins at different time courses after exercise in each group.The data were processed by SPSS 22.0 statistical software,and the data were expressed in the form of mean ± standard deviation,and the experimental results were analyzed by nonparametric test-rank sum test.Results:After one-time HIIT exercise,TNF in the aorta of rats first increased and then decreased,and a peak(P<0.05)appeared immediately after exercise,and the peak TNF expression in the adult group was higher.The expression of p21 first increased and then decreased,and the peak value(P<0.05)occurred immediately after exercise,the expression of Cyclin D1 decreased first and then increased,and the lowest value(P<0.05)appeared 3h after exercise.The expression of P4Hα1 decreased and then increased,and the lowest value(P<0.05)appeared 3h after exercise,the expression of MMP-2 first increased and then decreased and then increased,and the peak value(P<0.05)appeared immediately after exercise,lowest value appeared 12h after exercise.Conclusion:One-time HIIT exercise can induce inflammatory response,secrete more inflammatory factors,affect the level of cell cycle related factors,and then increase the expression of MMP-2 protein in aortic vessels,resulting in changes in collagen metabolism in aortic tissue.TNF-α,P4Hα1 and MMP-2 were more sensitive to the effects of disposable HIIT in adult rats during the experiment,suggesting that adult rats were more sensitive to disposable HIIT than young rats and more susceptible to the stimulation of disposable HIIT.
