| Research purpose:In this study,we observed the production of ketone body in the liver of mice after 6weeks of high-intensity interval training(HIIT)intervention and the expression of m TORC1,PPARα,FGF21 and other proteins closely related to ketone body production,and explored the effect of HIIT on ketone body production and its relationship with m TORC1/PPARα/ FGF21.Research methods:Twelve 7-week-old C57BL/6J male mice were randomly divided into control group(n=6)and HIIT group(n=6).Six weeks of formal HIIT training will be carried out after one week of exercise preconditioning: after 10 minutes of warm-up exercise(at a speed of 10 m/min),10 groups of high-intensity exercise will be carried out,each group of 6 minutes,including 4 minutes of high-intensity exercise(at a speed of 80-90% of the maximum speed)and 2 minutes of recovery exercise(at a speed of 40-50% of the maximum speed),with a platform slope of 10 °;1 time/day,5 days/week.At the end of adaptive training,at the end of the second week of training,and at the end of the fourth week of training,the maximum exercise speed that the mice can bear was evaluated,and the speed of high-intensity interval training was adjusted.The mice were weighed weekly and their weight was recorded.After 6 weeks of HIIT,the body composition of the two groups of mice was tested.Before the last exercise,immediately after the exercise and 1h after the exercise,blood was collected from the tail vein to detect the level of blood ketone body in mice.After the intervention,the mice were killed under anesthesia and the liver tissue was stripped and the serum was extracted.Extract liver protein for Western blot test,measure CPT-I,HMGCS2,HMGCL,βDH1,MCT1,MCT2,m TORC1,PPARα,FGF21 expression of protein.The expression of key enzyme genes HMGCS2,HMGCL and βDH1 for ketone body production was detected by PCR,and the level of free fatty acids in the liver and serum of mice was detected by the kit.Research results:1.The weight of mice in the control group increased gradually within 6 weeks,while the weight of mice in the HIIT group did not increase significantly.Compared with the control group,the body fat ratio of HIIT group mice decreased significantly(p<0.01),and the proportion of lean body weight increased significantly(p<0.01).2.Compared with that before HIIT,the blood ketone body level of mice immediately after HIIT and 1h after HIIT increased significantly(p<0.001,p<0.05),and the blood ketone body level of mice 1h after HIIT decreased compared with that immediately after HIIT.3.Compared with the control group,the level of free fatty acids in liver of mice after 6 weeks of HIIT increased significantly(p<0.05).4.Compared with the control group,HIIT at 6 weeks significantly increased the CPT-I and PPARα,FGF21 protein expression of mouse liver(p<0.01,p<0.01,p<0.05),the HMGCS2,HMGCL,βDH1 protein expression and gene expression of increased but not significantly(p>0.05),while the protein expression of m TORC1 decreased significantly(p<0.01).Research conclusion:HIIT can promote the production of ketone body in mouse liver,m TORC1/PPARα/FGF21 is its possible mechanism. |
