| Essential hypertension(EH) is a polygene trait and its occurrence and development are results of complex interaction between a variety of genetic and environmental factors. There is strong evidence that the renin-angiotensin system(RAS) plays an improtant role in the pathogenesis of EH and its complications. However, existing data from case-control studies of the association of ACE I/D polymorphysm and AGT M235T variant with blood pressure are conflicting,and up to date the exect associatiaon between gene variants of RAS and EH haven't established yet.It's well known that Tibetan people who living in Tibet are relatively isolated and genetically homogenous population with 22% of prevalence rate of hypertension,therefore it's very important to make pedigreee analysis and case-control study to observe familial aggregation ,heritability and metabolic characteristics of EH in Tibetan population,and it also deserve to exam the association of the genes of RAS and EH in this population. Materials and MethodsThe study was conducted in the Xue community and Duilong county of LHASA city and 53 EH pedigrees and 10 normotensive pedigrees were recriuted .106 unrelated EH patients and 135 normotensive subjects were also randomly recriuted from those two area.PCR,PCR/RFLP and PCR-SSCP were carried out to study the association of genes of RAS and EH . Results1. 44.06% first-degree relatives of EH pedigree are affected ,much higher than that of general population in Tibet. The heritability of EH was 85.3±10.33%.2. The fasting serum total cholesteral (TC) and serum triglyceride (TG) levels were significantly higher in hypertensive group of EH pedigrees(HF) than in normotensive group of EH pedigree(NF), non-direct relatives of EH pedigree(SN) and direct relatives of normotensive control group(CO),p<0.05.
