Objective:Ovarian cancer is the most common malignant tumor in women,in China,it ranks the third morbidity and the first mortality among the genital system cancer.Traditional Chinese Medicine(TCM)plays an important role either in suppressing,killing tumor cells or recuperating patients after operation,therefore it also helpful both in alleviating the toxic side-effect of chemoradiotherapy,and improving the quality of patients’ life.Consequently,the rearrangement,exploration and research of ancient medical conceptions and academic ideas in ancient books present significant modern medical value.Currently,the epitome of TCM,especially its feature of science and theory mostly depends on the academic thinking and clinical practice of the old medical experts.So we explored the medication rules and academic thoughts of Professor Shen Min-he in treating ovarian cancer,hoping to provide clinical supervision.Furthermore,to summarize some herbs which been used much frequently could provide formula-selection evidence for next part of the experiment.GSK-3β has an evident progression role in the life process of tumor cells,which makes it as a potential target for cancer therapy.Chinese herbal compound,which consists of several commonly used herbs in Professor Shen’s clinical practice has been chosen,and it’s underlying molecular mechanism of ovarian cancer metastasis inhibition by intervening GSK-3β has been investigated,for providing the experimental evidences in further improving the therapeutic efficacy of herbs.Methods:The first part:In this paper,the methods of literature research and theoretical review are applied.Take the time as a clue,through literature search and data identification,collection,research.To summarize ovarian cancer from 3 parts,such as the origin and evolution of the name,the description of the symptoms,and the etiology and pathogenesis.To figure out and sum up the cognitive rules and academic ideas of ancient physicians from successive dynasties.The second part:2014.1.1-2016.12.31,ovarian cancer outpatients who treated by professor Shen in Zhejiang Province Hospital of TCM has been recruited.Database established through data standardization processing.Descriptive analysis of the property and flavor,the channel entry,the classification of the most frequently used herbs(n=48)achieved by Excel2007 software.The cluster analysis of these highest frequent herbs(n=48)were made by SPSS19.0.The third part:After identified by the TCM formula preparation room,the production of Compound Fuling Granules was completed.The Hey and SKOV3 cells were cultured and induced by TGF-β to establish EMT model.The experimental techniques,such as proliferation test in vitro,Soft AGAR test,cell senescence test,nick experiment,Tran swell,Western Blot were used to investigate the correlation between the mechanism of Compound Fuling Granules inhibiting ovarian cancer cell EMT and Akt/GSK-3β signaling pathway.Results:The first part:The first sight into cancer could be traced back to Yin and Zhou Dynasties,according to the ancient literature,most physicians believe that ovarian cancer belongs to such kinds of disease category,that is "mass","Chang Xun" and " abdominal accumulation"and so on.The pre-Qin and Han Dynasties period was the initial formation stage of TCM theory.The classics "emperor’s Canon" and "Golden chamber" were cornerstone for Chinese physicians to treat "accumulation and mass".Northern and Southern Dynasties characterized by gradually developing towards specialization,physicians Wang Shuhe and Ge Hong described women "mass" in details.In the Sui and Tang Dynasties,with the thriving of evidence-based medicine(EBM),physicians paid more attention to the etiology and pathogenesis,treatment-differentiation of the single disease,and gradually formed a theoretical system.While the Song Dynasty had it’s own independent clinical discipline-Department of Obstetrics and Gynecology,and reached to a more comprehensive understanding of the etiology and pathogenesis,syndrome differentiation and prognosis in women "mass and accumulation".In Ming and Qing Dynasties,gynecology development into a unprecedented stage,a considerable amount of gynecology monographs completed.Then the late Qing Dynasty and the Republic of China,with the sprung of many periodicals of TCM and re-publication of classics,the tide of "integration of TCM and Western medicine"emerged.After the founding of new China,a comprehensive treatment mode of integrated TCM and Western medicine was adopted to ovarian cancer.The second part:The formula of Professor Shen was essential and simple,most prescription was compose of 7 to 12 herbs.The highest frequent herb was Poria cocos.The top 48 kinds of high-frequency herbs included by some prescription,such as guizhifuling pill、compound Fuling Tang(self-created)etc.The majority frequency distribution of syndrome pattern was deficiency syndrome,the other characteristics of frequency ’distribution were:warm in property;sweet,bitter,pungent in tastes;spleen prediction in channel chose;nourishment herbs in classification.Professor Shen proposed that the basic pathogenesis of ovarian cancer was "deficiency",complicated with,"cold","blood stasis","dampness".Therefore,methods of treatment "tonifying,warming and scattering","promoting blood circulation","eliminating dampness" were used in clinical practice.In addition,"Yin Yang and four seasons","modification of flavor and property","preserving of spleen and stomach","mental adjustment","compatibility and incompatibility of drugs" were also been emphasized.The third part:The study showed that AKT/GSK3β signal pathway was not only responsible to the inhibition of EMT,but also regulating the proliferation suppression and aging procession of biological functions.It concluded that the multi-target effect on ovarian cancer of Compound Fuling Granules’s may through regulating the AKT/GSK3 signal pathway.
