Objective:We will conduct data mining on professor Huang Yongsheng’s treatment of atrial fibrillation,and summarize the academic thoughts and medication rule of Professor Huang Yongsheng’s treatment of atrial fibrillation.Conduct network pharmacology research on the core prescription obtained from data mining,analyze the mechanism of the core prescription,and the mechanism of action was preliminarily verified and explained through molecular docking technology,in order to provide scientific basis for the clinical practice of TCM treatment of atrial fibrillation.Methods:(1)Collect and sort out the medical records of atrial fibrillation patients who were admitted to Professor Huang Yongsheng’s studio from January 1,2010 to December 31,2021.After screening according to the diagnosis,inclusion and exclusion criteria,the information of 206 medical cases was extracted into Excel and introduced into the cloud platform system of ancient and modern medical cases.With the help of statistical analysis,association analysis,systematic clustering,complex network analysis and other functions,the medical records were excavated from multiple angles,combined with professor Huang Yongsheng’s relevant works and daily clinical guidance,the academic thoughts and medication rule of Professor Huang Yongsheng’s diagnosis and treatment of atrial fibrillation were summarized.(2)To screen and collect the targets of the major components and effects of the core prescription of atrial fibrillation with gas deficiency and blood stasis syndrome.The relevant targets of atrial fibrillation were searched in the GeneCard database,OMIM database.The intersection of the two gets the therapeutic target.Build the "drug-component-target network through the software Cytoscape.A protein interaction network map was constructed through the String database and screened for core targets.The GO function analysis and KEGG analysis of the therapeutic targets were then conducted through the Metascape database.Finally,molecular docking technology was used to verify the binding effect of drug active ingredients and targets.Results:(1)Data mining research①Of the 206 cases,113(54.9%)were male and 93(45.1%)were female;spanning 24 to 92 years,with the highest incidence from 60 to 79 years(59.73%).②There were 73 symptoms In 206cases,and the top 10 symptoms were fatigue,palpitations,chest tightness,shortness of breath,upset,excessive breath,dry mouth,dizziness,fear of cold and insomnia.In the tongue image,there are 9 kinds of tongue,3 kinds of tongue body and 11 kinds of tongue moss,and the most common are blue and purple tongue,tooth marks,tongue and thin moss;There are 14 kinds of veins,and the first three kinds of veins with the most frequency are pulse formation,pulse heavy string thin and pulse string heavy.③ Diagnosis:The most complicated diseases were coronary heart disease,hypertension,chronic heart failure,diabetes,and lacunar cerebral infarction;there were 9 syndrome types,and the top three were 164(79.61%)of qi deficiency and blood stasis,15(7.28%),and 14(6.80%).④Prescription Information:A total of 206 cases of Chinese medicine used 154 flavors,frequency of the top 10 Chinese medicine:Danshen、Sharen、Huangqi、Kushen、Guizhi、Jiangxiang、Suanzaoren、Gancao、Baishao、Dazao.The traditional Chinese medicine is the most beneficial;The drug nature is mainly warm drugs,the five flavors are mainly sweet and spicy traditional Chinese medicine,and the drugs belong to the liver meridian,spleen meridian,stomach meridian is the most.Support was set to 0.7 and confidence set to 0.9,yielding 31 groups of associated drug pairs.Hierarchical clustering analysis of TCM yielded two combined results,combination 1:Suanzaoren、Cishi、Zhiqiao、Qingpi、Chenpi、Cangzhu、Baizhu;combination 2:shuizhi、Jiangcan、Chantui、Dangshen、Jiangxiang、Baishao、Gancao、Guizhi、Dazao、Kushen、Huangqi、Danshen、Sheran.⑤Statistical results of medical cases of qi deficiency and blood stasis group:The top 10 traditional Chinese medicines of use frequency are Haungqi、Danshen、Sharen、Kushen、Guizhi、Ganccao、Dazao、Baishao、Suanzaoren、Jiangxiang.Support was set to 0.4 and confidence to 0.9,obtaining 30 groups of associated drug pairs;Hierarchical clustering analysis yielded 3 sets of drug compatibility combinations,combination 1:Jaingxiang、Suanzaoren;combination 2:Dazao、Gancao、Guizhi、Baishao;combination 3:Danshen、Sharen、Kushen、Haungqi.⑥ Statistical results of medical cases of sputum and blood stasis mutual group:The top 10 traditional Chinese medicines of use frequency are Zexie、Danshen、Yinchen、Fuling、Sharen、Qingpi、Shichangpu、Yujin、Zhiqiao、Zhuru;Support was set to 0.2 and confidence to 0.6,yielding 16 groups of associated drug pairs.Hierarchical clustering analysis yielded 4 sets of drug compatibility combinations,combination 1:Chenpi、Zhuru、Zhiqoiao、Yujin、Shichangpu、Qingpi、Sharen;combination 3:Yinchen、Danshen、Zexie、Fuling、Kushen;combination 3:Baizhu、Cangzhu、Houpo、Tanxiang;combination 4:Jiangxing、Shuihzi、Chantui、Jiangcan.⑦ Statistical results of the medical cases of qi and Yin two deficiency group:The top 10 traditional Chinese medicines of use frequency are Huangqi、Qingpi、Fuling、Danshen、Zhiqiao、Shengdihuang、Shudihuang、Cangzhu、Shanyao、Guizhi.Support was set to 0.3 and confidence to 0.8,yielding 15 associated drug pairs;Hierarchical clustering analysis yielded 3 sets of drug compatibility combinations,combination 1:Suanzaoren、Shouwuteng、Kushen、Shanyao、Shanzhuyu、Gouqizi;combination 2:Gancao、Baishao、Guizhi、Dazao;combination 3:Sharen、Baizhu、Cangzhu、Danshen、Fuling、Shengdihuang、Shudihuang、Huangqi、Qingpi、Zhiqiao.4(2)Network pharmacology and molecular docking studiesA total of 277 targets were obtained from the core prescription to target 1,187 atrial fibrillation disease targets,and 107 therapeutic targets were obtained after taking the intersection;Four core active components were obtained:polyquercetin,luteolin,β-glusterol,kaempferol,and five key targets:AKT1、TNF、IL6、VEGFA、IL1B;The GO functional analysis yielded 209 biological processes,84 cellular components,and 94 molecular functions;The KEGG analysis yielded 194 signaling pathways.The results of molecular docking experiments showed that four major active components bind stably to five key targets.Conclusion:When treating atrial fibrillation,Professor Huang Yongsheng pays attention to the overall syndrome differentiation,takes Yin and Yang as the general principle,first distinguishes the deficiency and deficiency of the viscera,and pays attention to the influence of qi stagnation,phlegm turbidity and blood stasis,and believed that the disease of atrial fibrillation is in mind,but it also points out that "the treatment of the heart is not idealistic".Therefore,in clinical practice,different methods of atrial fibrillation,such as the treatment of fear,adjustment,adjustment of liver and kidney fear,stabilization and control,and stabilization,are summarized.According to data mining,professor Huang Yongsheng should make good use of qi and blood circulation drugs to invigorate qi and blood circulation throughout,replenishing qi and blood circulation,and added ginseng,prince ginseng,red ginseng,Party ginseng and Korean ginseng;Blood circulation medicine with Danshen drink as the basic prescription,burning Panax notoginseng and zedoary,blood stasis is heavier,add cicada,stiff silkworm,leeches,and even more with heavy agent to promote blood circulation medicine Panax notoginseng and the core prescription could act on the targets of AKT1,TNF,IL 6,VEGFA,IL1B through polyquercetin,luteolin,β-sitosterol,kaempferol and other constituents.In addition,MAPK signaling pathway,PI3 PK-Akt signaling pathway,relaxin signaling pathway,TNF signaling pathway were used to inhibit inflammatory response,improve myocardial fibrosis,regulate ion channels,and regulate apoptosis,reflecting the characteristics of multi-components,multi-targets and multi-pathway to treat diseases. |
