| Objective:1.With the help of "Traditional Chinese Medicine Inheritance computing Platform(V2.5)",this study analyzed and mined the clinical medication rules of traditional Chinese medicine in the treatment of female drench syndrome,deeply understood the connotation of drug compatibility,and explored new combinations of potential drugs to provide more data support for the clinical treatment of female drench syndrome.2.Using network pharmacology to predict the underlying substance basis and mechanism of action of high-frequency drug therapy for urinary tract infection at molecular level.Methods:1.Data mining: From January 1,2011 to December 31,2020,the clinical literatures on TCM treatment of female gonorhea were searched in the online databases CNKI,Wanfang database and VIP.178 literatures were obtained according to the literature inclusion standards.After the drug names were processed in a standardized way,Import it to "TCM Inheritance Computing Platform" for data mining analysis,and use frequency statistics,association rule analysis and cluster analysis to mine out high-frequency drugs,core drugs,high-frequency drug pairs and new prescription combinations.2.Using the method of network pharmacology,the components and targets of high-frequency drugs extracted in the early stage of traditional Chinese Medicine inheritance computing platform were screened by TCMSP.The gene targets of urinary tract infection were screened by Drugbank,Gene Cards and OMIM databases.After the protein gene standardization of drug and disease targets was carried out by Uniprot,VENNY2.1 online tool was used to draw Venn diagram to obtain common targets of drugs and diseases,PPI network analysis was performed on common targets through STRING database,common targets were imported into Metascape database,species were set as human,GO functional annotation and KEGG enrichment analysis were performed on common targets.The top 20 results with the smallest P value in GO and KEGG results were visualized.Results:1.Data mining results: Drug frequency: The top 10 drugs in clinical treatment of female gonorrhea were glycyrrhiza,Poria cokahoe,Cortex quinqueae,Phellodendron phellodendron,Bupleurum,polygonum polygonum,Radix rehmanniae,Plantain fructus,Yam and Alisma orientalis.The four qi of drugs were cold,1298 times,accounting for 48.32%.Flat,651 times,24.24%;Temperature,584 frequency,accounting for 21.74%;Coolness,102 times,3.8%;Thermal,51 times,accounted for 1.9%.The five flavors were sweet,1629 times,accounting for 41.42%;Bitter taste,1267 times,32.21%;Spicy taste,657 times,accounting for 16.7%,sour taste,203 times,accounting for 5.16%;Astringency,113 times,accounting for 2.87%;Saltiness,64 times,accounting for 1.63%.The results of meridian regression were liver meridian,1157 times,accounting for 15.83%;Kidney meridian,1058 times,accounting for 14.48%;Spleen meridian,960 times,accounting for 13.14%;Lung meridian,960 times,accounting for 12.52%;Gastric meridian,875 times,accounting for11.97%;Cardiac meridian,850 times,accounting for 11.63%;Bladder meridian,532 times,accounting for 7.28%;Small intestine meridian,358 times,accounting for 4.90%;Large intestine meridian,317 times,accounting for 4.34%;Biliary meridian,218 times,accounting for 2.98%;Pericardial channels,45 times,accounting for 0.62%;Tricoke meridian,22 frequency,accounting for 0.30%.Through association rule analysis,a total of 92 drug combinations were obtained,including 26 TRADITIONAL Chinese medicines,and the confidence degree of 58 rules was ≥0.60,and 5 core drugs were obtained.A total of 9 new methods were excavated by cluster analysis.2.A total of 173 active components,including quercetin,luteolin,kaangferol and naringin,were predicted based on network pharmacology for the treatment of urinary tract infection.After removing repeated targets,there were 271 targets of high-frequency drug active ingredients,and 147 potential targets corresponding to drugs and diseases,including AKT1,TNF,TP53,IL6,JUN,etc.GO biological function enrichment analysis results: In biological process,mainly reflected in reaction to inorganic substances,cell reaction to nitrogen compounds,cell reaction to lipids,response to external stimuli,etc.Cell composition involves membrane valve,vesicle cavity,extracellular matrix and many other locations.Target genes exert biomolecular functions mainly by regulating protein homologous dimerization activity,DNA binding transcription factor,nuclear receptor activity,kinase binding,REDOX enzyme activity and protein kinase activity.Furthermore,KEGG pathway enrichment analysis revealed that these proteins may regulate UTI pathological process through MAPK signaling pathway.Conclusions:1.The basic principles of clinical treatment of female drench syndrome are as follows:relieving dampness of water as the core,supplementing heat clearing and deficiency tonic,indicating that female drench syndrome is based on kidney deficiency and bladder dampness and heat as the standard.Common high frequency drugs used to treat female drenching syndrome are licorice,Poria cocos,Rhizoma quumata,Phellodendron phellodendron,Bupleuri,polygonum polygonum and rehmannia sativa.2.Data mining of high-frequency drugs by quercetin,mignonette,galangal,pomelo,mans,handle,active ingredient by diseases such as licorice A chalcone targets AKT1,TNF,TP53,IL6,JUN,such as function,affect multiple biological effects,mediated the activation of MAPK signal pathway and conduction,urinary tract infection in pathologic process adjustment. |
